Type 1 Diabetes Mellitus (T1DM) is a complicated autoimmune disorder initiated by T-cell-mediated damage of pancreatic beta cells, resulting in insulin deficiency and the development of hyperglycaemia. This disease is most common in childhood and adolescence and frequently co-occurs with other autoimmune conditions like autoimmune thyroiditis. This work aimed to investigate thyroid autoantibodies and their correlation with thyroid functions and tumour necrosis factor-alpha (TNF-α) in T1DM patients. Eighty participants were enrolled in a case-control study, including sixty T1DM patients and twenty healthy controls. Peripheral blood specimens were taken from individuals with proven T1DM and healthy individuals (control group). Body mass index (BMI), fasting blood sugar (FBS), glycated hemoglobulin (HbA1c), thyroid-stimulating hormone (TSH), free triiodothyronine (FT3), free thyroxine (FT4), thyroid autoantibodies, including anti-thyroid peroxidase (anti-TPO) in addition to anti-thyroglobulin (anti-Tg), and TNF- α concentrations were evaluated in T1DM and control groups. Our findings revealed that thyroid autoantibodies were significantly more prevalent in T1DM patients, elevated TSH, and significantly higher TNF-α levels than in the control group, indicating an increased risk for inflammation based on cytokine levels. The current study proved that in T1DM patients' sera compared to the healthy control, thyroid autoantibodies and TNF-α levels were positively correlated. In conclusion, poor diabetes management and high TSH levels may indicate subclinical hypothyroidism, which impacts diabetes control. This is possibly linked to thyroid autoimmunity. Future research is needed to understand how TNF- αcontributes to the progression of diabetes disease and its complications.

Interleukins (ILs) are a major subclass of cytokines which act as molecular messengers playing a role in immune system responses via cascade of signalling pathways. Belonging to the cytokine family, ILs play a crucial role in the theronostics of various diseases. Their abnormal expression leads to development of various diseases such as cancer, neurodegenerative diseases, allergies, asthma, autoimmune diseases, and other physiological abnormalities. This paves the way for exploring ILs to develop sensitive and efficient biosensors, and promoting them for clinical testing in a wide array of diseases. Furthermore, detecting the level of ILs is very important for their early diagnosis and their progression within the body, and simultaneously for possible immunotherapeutic approaches. To achieve this goal, multidisciplinary scientific approaches involving immunology, electrochemistry, nanotechnology, photometry, etc. are already being put into action. Advancements in nanoscience and nanotechnology are aiding in the development of highly sensitive biosensors for ILs detection. This review focuses on a detailed description of all presently discovered ILs and their roles in various diseases. Simultaneously, it also discusses the various electrochemical biosensors that can be employed for the detection of ILs in bodily fluids. Moreover, the role of nanomaterials in electrochemical biosensing are also discussed here.

Exploring the Potential Link between Vitamin D Receptor Cdx2 Gene Polymorphisms and Rheumatoid Arthritis in Female Patients: A Study in Erbil, Iraq

Background: In the last decades, vitamin D has been a subject of research in several autoimmune diseases, including Rheumatoid Arthritis (RA), a chronic inflammatory disease characterized by a distinctive pattern of joint involvement and joint destruction. Aim: The main goal of the study is to examine the relationship between (RA) and the polymorphism in the vitamin D receptor's Cdx2 gene in a sample of women. Methods: Eighty females (60 with RA and 20 controls) participated in this case-control investigation. The Cdx2 single nucleotide polymorphisms (SNP) of the VDR gene were genotyped using tetra-primer amplification refractory mutation system-polymerase chain reaction technique (ARMS-PCR). Results: The current study discovered that 51 RA patients have the heterozygous GA genotype of the Cdx2 polymorphism despite non-significant values. Although the present findings showed no significant association between Cdx2 gene polymorphisms and the risk of RA, this genotype still represents the majority among the study's RA patients. Moreover, the results showed that the serum VD3 was significantly lower in RA with the GA genotype compared to healthy controls with a similar genotype (p=0.0002). Also RA patients with heterozygote GA genotype for Cdx2 had a relatively lower mean for serum vitamin D3 level than those with homozygote GG genotype. Conclusion: The novelty of this study is to discover the probable association of Vitamin D receptor, caudal-related homeobox 2 (VDR-Cdx2) polymorphism with RA in the given study group for the first time in our society using a tetra primer ARMS-PCR. Additionally, the tetra-primer ARMS-PCR technique was simple, reliable, low-cost, and might be a valuable substitute for Restriction fragment of length polymorphism- polymerase chain reaction– (RFLP-PCR) in routine clinical settings for single nucleotide polymorphism (SNP) genotyping.
