Corneal injury: Clinical and molecular aspects

Abstract: Currently, over 10 million people worldwide are affected by corneal blindness. Corneal trauma and disease can cause irreversible distortions to the normal structure and physiology of the cornea often leading to corneal transplantation. However, donors are in short supply and risk of rejection is an ever-present concern. Although significant progress has been made in recent years, the wound healing cascade remains complex and not fully understood. Tissue engineering and regenerative medicine are currently at th… Show more

“…83,84 Several other gene knockdown strategies for ocular scarring are also being tested in animals. 1,[9][10][11]14 In this study, we used the fully modified siRNA conjugate to achieve maximal activity and longevity of the effect in vivo. 48 Regarding the possibility of off-target effects, the asymmetric (20/15) chemically modified siRNA used in this study was designed to avoid any sequence identity with other rabbit genes within the 2-18 region of the anti-sense strand.…”

“…Corneal scarring results from mechanical injury, burn, infection, or surgery. 1 Other examples of ocular scarring include glaucoma filtration surgery (the bleb to relieve intraocular pressure can heal fibrotically) 2 and other ocular morbidity such as proliferative vitreoretinopathy (PVR) and retinal detachment. 3,4 Mitomycin C (MMC) to improve healing and avert scarring is a standard of care for some of these indications, but there is a high failure rate with the filtration surgery and cell toxicity concerns in corneal surgeries.…”

“…Keratoconus (KC) is an asymmetrically bilateral corneal dystrophy characterized by progressive thinning of the cornea, which becomes cone-shaped in a process called ectasia, with a prevalence of approximately 1 in 2000 people [1][2][3][4][5][6][7]. It is currently known that KC is a multifactorial disease, involving genetic, environmental, and behavioral factors and etiology remains poorly understood [3,[7][8][9][10][11][12][13][14].…”
