Cord–Blood–Derived Human Cultured Mast Cells Produce Interleukin 13 in the Presence of Stem Cell Factor

Abstract: Background: Mast cells have been regarded as a potential source of cytokines. Although the human mast cell line HMC–1 and human lung mast cells have been shown to produce interleukin (IL) 13, it still remains uncertain whether cord–blood–derived human cultured mast cells produce IL–13. Methods: Human cultured mast cells were raised from cord blood cells in the presence of stem cell factor (SCF) and IL–6. Levels of IL–13 mRNA were examined by a semiquantitative reverse transcriptase–polymerase chain reaction. I… Show more

“…It has been implicated in the induction of airway hyperresponsiveness and mucus hypersecretion (37,38). SCF may play an essential role in the regulation of IL-13 production because anti-IgE-induced IL-13 production by human mast cells is dependent on SCF (39). In addition, inhibition of SCF induced decreased airway hyper-responsiveness (40).…”

“…Total cellular RNA was extracted by the method of acid guanidinium thiocyanate (GT)-phenol-chloroform extraction [27,28]. Oligonucleotide primers were synthesized by Kurabou Co. (Kurashiki, Japan) and their sequences are summarized as follows; p140 trk primers were 5 H -TGGAG AAGAA GGACG AAACA-3 H (sense, located at nucleotides 1277±1296) and 5 H -GCCTT GACAG CCACC AGCAT-3 H (antisense, at 1669±1688); p75 LNGFR primers, 5 H -GACAG TGGCA TCTCC GTGGA CA-3 H (sense, at 1023±1044) and 5 H -ACACC GGGGA TGTGG CAGTG GA-3 H (antisense, at 1374±1395).…”

“…SCF has been shown to induce hematopoietic cell activation and interleukin (IL)-13 and IL-6 release (21,22). In the present work, OD-like cells were found to produce SCF after contact with a bacterium-derived antigen.…”
