Coordination between Rac1 and Rab Proteins: Functional Implications in Health and Disease

Abstract: The small GTPases of the Rho family regulate many aspects of actin dynamics, but are functionally connected to many other cellular processes. Rac1, a member of this family, besides its known function in the regulation of actin cytoskeleton, plays a key role in the production of reactive oxygen species, in gene transcription, in DNA repair, and also has been proven to have specific roles in neurons. This review focuses on the cooperation between Rac1 and Rab proteins, analyzing how the coordination between thes… Show more

“…An alternative possibility is that EIPA inhibits VSVΔG-BHLD entry due to its previously described pleiotropic effect manifested as gross reorganization of the endosomal network and changes in Na + and H + gradients in the cell (71, 72). Similarly, inhibition of Rac1 by NSC23766 could have a pleotropic effect because Rac1, in addition to its roles in regulating the actin cytoskeleton, is also involved in several other cellular processes (73).…”

“…EIPA treatment can lead to a gross reorganization of the endosomal network and changes in Na + and H + gradients in the cell (94, 95). Similarly, Rac1, the target of NSC23766, is involved in several cellular processes in addition to regulating the actin cytoskeleton (96).…”

“…The expression pro les of ARLs are rstly explored using TCGA, GTEx and Oncomine databases, which demonstrate that ARLs are commonly dysregulated in GC. Accumulating evidences suggest that the cross-talk and collaboration between small GTPase proteins are causatively involved in several cellular processes and diseases (24)(25)(26)(27). In this study, co-expression and correlation analyses demonstrate that the expression levels of ARLs in GC show high correlations.…”

“…Therefore, an attractive mechanism for the timely remodeling of local actin cytoskeletal networks in concert with dynamic changes of membranes may be efficient coordination of Rho and Rab GTPases. Indeed, evidence is emerging that reveals an interplay between Rab and Rho GTPases [8,9].…”
