Cooperation of Angiopoietin-2 and Angiopoietin-4 in Schlemm's Canal Maintenance

Abstract: Purpose Defects in the iridocorneal angle tissues, including the trabecular meshwork (TM) and Schlemm's canal (SC), impair aqueous humor flow and increase the intraocular pressure (IOP), eventually resulting in glaucoma. Activation of endothelial tyrosine kinase receptor Tie2 by angiopoietin-1 (Angpt1) has been demonstrated to be essential for SC formation, but roles of the other two Tie2 ligands, Angpt2 and Angpt4, have been controversial or not yet characterized, respectively. Met… Show more

“…Similar to Ang1, and unlike Ang2, Ang4 induces Tie2 autophosphorylation upon binding to Tie2 [52]. Ang4 appears to play a role in venous development of the mouse retina and in aqueous humor drainage [52,53].…”

“…Angpt2 gene deletion is compatible with embryonic vascular development but subsequently results in abnormal angiogenic remodeling and defective lymphatic vessels (22). Deletion of the third angiopoietin (Angpt4) in mice results in defective venous maturation in the retina (23), and combined deletion of Angpt2 and Angpt4, in a hypomorphic Schlemm's canal (lymphatic-like vessel in the anterior part of the eye) and glaucomatous changes in the eyes (24).…”
