Converse Smith-Martin cell cycle kinetics by transformed B lymphocytes

Abstract: Recent studies using direct live cell imaging have reported that individual B lymphocytes have correlated transit times between their G1 and S/G2/M phases. This finding is in contradiction with the influential model of Smith and Martin that assumed the bulk of the total cell cycle time variation arises in the G1 phase of the cell cycle with little contributed by the S/G2/M phase. Here we extend these studies to examine the relation between cell cycle phase lengths in two B lymphoma cell lines. We report that t… Show more

“…Cell cycle period and phase durations of mammalian cells demonstrate a high degree of heterogeneity under culture conditions and within a tumor microenvironment. , Such heterogeneities often significantly influence cell fate decision-making. This means that these heterogeneities can be fine-tuned to design better therapeutics if the factors controlling these variabilities are known precisely. However, identifying these controlling factors is highly challenging due to the diverse and complex nature of cell cycle duration heterogeneities.…”

“…Identifying the governing factors that organize the cell duration heterogeneities is a challenging task (8,9,12,17,(19)(20)(21)(22). In this article, we combined live-cell imaging studies of FUCCI-HeLa cells (Fig.…”

“…For example, The cell cycle phase durations quantified at the single-cell level in proliferating lymphocytes (8,17) using FUCCI-reporter (18) showed a highly variable S-G2-M time with strongly correlated G1 timings in the sibling cell pairs. A highly correlated cell cycle in sibling pairs has been observed in other mammalian cell types as well (19)(20)(21)(22) with mothers and daughters having poor correlation in cell cycle timings, while cousins showing a significant correlation for the same (19)(20)(21)(22).…”
