Contribution of serotonin uptake and degradation to myocardial interstitial serotonin levels during ischaemia‐reperfusion in rabbits

Abstract: The 5-HT reuptake function plays an important role in the clearance of myocardial interstitial 5-HT during reperfusion. When 5-HT reuptake function is intact, degradation of 5-HT by monoamine oxidase contributes to reduce myocardial interstitial 5-HT level throughout ischaemia-reperfusion.

“…The materials and properties of the dialysis probe have been described previously (Sonobe et al, 2013(Sonobe et al, , 2014Kawada et al, 2012Kawada et al, , 2013. Briefly, each end of a dialysis fiber (0.31 mm O.D., 0.2 mm I.D., 50,000 molecular weight cut off; PAN-1200, Asahi Chemical, Japan) was inserted into a polyethylene tube (25 cm length, 0.5 mm O.D., 0.2 mm I.D.)…”

“…After baseline sampling, one dialysis probe was perfused with Ringer solution containing pargyline (1 mM) and the other with Ringer solution containing fluoxetine (1 mM), continuously for 120 min. The concentrations of pargyline and fluoxetine were determined based on our previous cardiac microdialysis studies (Yamazaki et al, 2001;Sonobe et al, 2013). Sixty minutes after administration was started, dialysate was sampled from each dialysis probe continuously for 60 min (three 20-min samples) (Fig.…”

“…The first dialysate sampling was started from at least 2 hr after the probe implantation. Serotonin concentration in the dialysate sample was measured by electro‐chemical detection coupled with high performance liquid chromatography (HPLC‐ECD, Eicom) as described previously (Du et al, , ; Sonobe et al, ).…”

“…Serotonin (5‐hydroxytryptamine, 5‐HT) accumulates in the heart during cardiac ischemia (Du et al, ; Shimizu et al, ; Sonobe, Akiyama, Du, Zhan, & Shirai, ). The increased myocardial interstitial level of 5‐HT directly induces myocardial cell injury through 5‐HT receptor‐dependent coronary vasoconstriction (Golino et al, ; Métais, Bianchi, Li, Simons, & Sellke, ), platelet aggregation (Willerson, Golino, Eidt, Campbell, & Buja, ), and overactivation of afferent cardiac sympathetic nerves (Fu & Longhurst, ; Longhurst, Tjen‐A‐Looi, & Fu, ).…”
