Constituents of Machilus thunbergii bark and inhibition of cyclin-dependent kinases (CDKs) by procyanidin B2

Abstract: Stigmast-5-en-3β,7β-diol (1), stigmast-5-en-3β,7α-diol (2), (+)-isolariciresinol-9-O-β-D-xylopyranoside (3), procyanidin B2 (4), and epicatechin-(4β→8)-epicatechin-(4β→6)-epicatechin (5) were isolated for the first time from the bark of Machilus thunbergii. The chemical structures of the compounds were determined based on nuclear magnetic resonance, electronic ionization mass spectrometry, fast atom bombardment mass spectrometry, and IR. Compound 4 demonstrated inhibitory activity against cyclin-dependent kina… Show more

“…In 2011, five sterols from the leafy culms of D. bipinnata were reported and identified as stigmasterol, β-sitosterol, daucosterol, stigmast-5-en-3β,7β-diol and stigmast-5-en-3β,7α-diol [22]. A new xanthene was isolated from methanolic extract, which was identified as 2,6-dihydroxy-7-methoxy-3H-xanthen-3-one ( Figure 1) [23]. In 2009, flavonoid compound 4'-methoxy quercetin-7-O-glucoside ( Figure 1) was isolated from methanolic extract [24].…”

“…Some aurones with N-methylpiperidinyl at 7 position have been reported for inhibition of CDK1, CDK2 and CDK4 but not for CDK5/p25. 20 There are few natural products such as butyrolactone, 21 procyanidin B2, 22 hymenialdisine 23 and bellidin 24 that have been reported for CDK5/p25 inhibition. Recently, some effective CDK5/p25 inhibitory compounds including litvinolin, 6-O-galloyl arbutin, 4 0 -methoxyherbacetin, 6-O-galloyal arbutin, epigallocatechin-3-gallate, have been reported from Rhodiola rosea.…”
