Concentration Profiling in Rat Tissue by High-Resolution Magic-Angle Spinning NMR Spectroscopy:  Investigation of a Model Drug

Abstract: The utility of high-resolution magic-angle spinning (HR-MAS) NMR for studying drug delivery in whole tissues was explored by dosing female Sprague-Dawley rats with topical or injectable benzoic acid (BA). In principle, HR-MAS NMR permits the detection of both intra-and extracellular compounds. This is an advantage over the previous detection of topically applied BA using microdialysis coupled to HPLC/UV as microdialysis samples only the extracellular space. Skin and muscle samples were analyzed by 1 H HR-MAS N… Show more

“…Lunte and co-workers have published a number of papers [37,38] using CE and LC for MD separations, the same group also reviewed the area of MD with regards to analytical considerations [14]. Most recent published works [39] involved a RP separation using UV detection at 300, 280 and 275 nm to monitor the absorption of salicylic acid (SA), caffeine and metoprolol in the stomach following …”

“…High-spatial-resolution 1 H magnetic resonance spectroscopy (MRS) is a non-invasive method to profile metabolites (23). Compared with lower magnetic field strengths, ultra-high field strengths, including 7-Tesla (T) and 9.4T, provide a higher spectral resolution and signal-to-noise ratio, which significantly enhances the information obtained from these spectra.…”

“…5,12 Metabolite identification and quantification in a small volume (few microliters) of dialysate fluid are usually performed remotely from the patients or animals using high-sensitivity analytical techniques, such as chromatography, mass spectrometry or high-resolution nuclear magnetic resonance (NMR). [13][14][15][16][17] Despite their high detection sensitivity, the complexity of these techniques requires their application remotely from the subjects, leading to lengthy analytical protocols (tens of minutes to hours) and preventing the investigation of real-time metabolic changes in dialysate composition.…”
