Comprehensive Proteomic Characterization of the Pectoralis Major at Three Chronological Ages in Beijing-You Chicken

Abstract: Chronological age is one of the important factors influencing muscle development and meat quality in chickens. To evaluate the protein expression profiles during skeletal muscle development, we performed a tandem mass tag (TMT)-based quantitative proteomic strategy in pectoralis major (breast muscle) of Beijing-You chicken (BYC) at the chronological age of 90, 120, and 150 days. Each chronological age contained 3 pooling samples or 15 birds (five birds per pooling sample). A total of 1,413 proteins were identi… Show more

“…Thereafter, the peptide was labeled with TMT (Thermo Fisher Scientific, Waltham, MA), and the TMT-labeled peptides were fractionated on an Agilent 1290 HPLC operating at 0.3 mL/min using a Waters XBridge BEH130 column (C18, 3.5 μm, 2.1 mm × 150 mm). The fractions were dried for nanoliquid chromatography–tandem mass spectrometry (LC–MS/MS) analysis according to a previous study …”

“…To validate the TMT proteomics data, further liquid chromatography-parallel reaction monitoring MS (LC-PRM/MS) analysis was performed ( 30 ). PRM analysis was performed on a Q Exactive HF-X mass spectrometer (Thermo Fisher Scientific) and the optimized methods were generated experimentally using 1–3 unique peptides of high intensity and confidence for each target protein.…”

“…However, these studies have mainly focused on IMF ( 26 , 27 ) and abdominal fat ( 31 , 32 ). Most of them were limited on embryonic muscle ( 28 , 29 ) or during the bird body development stage ( 30 , 31 ). To date, there was no report on the dynamic changes in FA composition and related proteins in chicken during the laying period, especially from the sexual maturation stage to the culling stage.…”

“…Some of the annotated genes within the CNVR have been already associated to functional traits in chickens (Additional le 5). The CNVR located on chromosome 1, shared among different animals, were used to annotate several genes, the most noteworthy being CACNA2D1 (calcium voltage-gated channel auxiliary subunit alpha 2 delta 1) which is related to muscle contraction [41]; DACH1 (dachshund family transcription factor 1), involved in skeletal development and inhibitor of growth factor beta [42,43]; DMD (dysthrophin), one of the most important factors for muscle development and structural stability of the tissue [44]; and NTS (neurotensin) which is associated to neuronal network work ow [45]. Noteworthy, the gene encoding the miRNAs, MIR7445-1, is present within a mixed CNVR identi ed in several animals belonging to different breeds (Additional le 5).…”
