Complete Genome Sequence of Mycoplasma synoviae Strain WVU 1853 T

Abstract: A hybrid sequence assembly of the complete Mycoplasma synoviae type strain WVU 1853T genome was compared to that of strain MS53. The findings support prior conclusions about M. synoviae, based on the genome of that otherwise uncharacterized field strain, and provide the first evidence of epigenetic modifications in M. synoviae.

“…Among the CRISPR- associated genes, csn1 appeared intact in MS53, but was present as a pseudogene in WVU-1853 and MS-H. In WVU-1853, csn1 was disrupted by a single nucleotide deletion resulting in a frameshift [ 11 ]. In MS-H, csn1 was disrupted by an 11 nucleotide insertion.…”

“…Such knowledge would help to define molecular markers for tracking MS-H in vaccinated flocks and assessing any variation in the level of cross-protection against local strains of M. synoviae . The sequences of the M. synoviae field strain MS53, isolated around 2003 from a broiler flock in Brazil [ 10 ], and the type strain WVU-1853, isolated in 1957 from a hock joint of chicken in the USA [ 11 ], are the only two complete genomes determined thus far for this species. This is possibly due to difficulties experienced in assembling next generation sequencing (NGS) data for M. synoviae , which contains large, low complexity, repeat-rich regions.…”

“…Until now, the complete genomes of six strains of M. synoviae from four continents have been published, which can provide a preliminary understanding of the similarities and differences of different M. synoviae strains from the same and different geographic locations. ATCC 25204 (GenBank accession No: NZ_CP011096.1) and NCTC10124 (GenBank accession No: NZ_LS991953.1) are from the same type strain WVU1583, which was isolated from a chicken joint in West Virginia, USA (6,37). ATCC 25204 is stored in the American Type Culture Collection, while NCTC10124 is preserved in the Wellcome Trust Sanger Institute, United Kingdom.…”

“…The assembled genomes (18DW, 51SH, BS4S2, G3, and A4) and the genomes of MS-H [12] and WVU1853 [13] from GenBank were annotated using the Prokka, a genome annotation tool [14]. The average length of the assembled genomes was 795,375 bp.…”

“…Each MS isolated from farms in Korea from 2017 to 2020, MS-H vaccine (Vaxsafe MS, Bioproperties, Australia) and WVU1853 (ATCC 25204) were also used in this study. Fasta files from accession number CP021129 and CP011096 were used to retrieve the genomes of MS-H [12] and WVU1853 [13], respectively (Table 5). MFB with and without 0.01% NAD supplement (Sigma-aldrich, Saint Louis, MO, USA) were used to incubate the MS isolates from Korea, strain WVU1853 and MS-H vaccine.…”
