Complete assessment of whole-body n-3 and n-6 PUFA synthesis-secretion kinetics and DHA turnover in a rodent model

Abstract: Previous assessments of the PUFA biosynthesis pathway have focused on DHA and arachidonic acid synthesis. Here, we determined whole-body synthesis-secretion kinetics for all downstream products of PUFA metabolism, including direct measurements of DHA and n-6 docosapentaenoic acid (DPAn-6, 22:5n-6) turnover, and compared n-6 and n-3 homolog kinetics. We infused labeled α-linolenic acid (ALA, 18:3n-3), linoleic acid (LNA, 18:2n-6), DHA, and DPAn-6 as H-ALA, C-LNA, C-DHA, and C-DPAn-6. Eight 11-week-old Long Evan… Show more

“…EPA and DHA can also be synthesized from ALA [7], a process that involves several steps orchestrated by multiple elongases, desaturases, and β-oxidases [8]. However, the synthesis of EPA from ALA occurs at a low rate in mammals [9]. Of note, the same enzymes are employed by omega-6 fatty acids for their metabolic pathways.…”

“…However, in western countries DHA intake from fish consumption is very low compared to the intake of LA (from vegetable oils) and AA (from eggs and meat) [ 11 ]. In addition to the high dietary intake of n -6 fatty acids, the synthesis of AA from LA when this fatty acid is highly consumed is more efficient than the synthesis of DHA from ALA [ 12 ].…”

“…Domenichiello and Bazinet et al elaborated and widely applied this approach. Using this method, Domenichiello et al (Domenichiello et al 2014, Domenichiello et al 2016, Metherel et al 2018) estimated whole body synthesis-secretion rates of DHA from circulating ALA to be 1.5 and 0.045 μmol/day in rats consuming a high ALA or a low ALA containing diet, respectively.…”
