Competitive endogenous RNA is an intrinsic component of EMT regulatory circuits and modulates EMT

Abstract: The competitive endogenous RNA (ceRNA) hypothesis suggests an intrinsic mechanism to regulate biological processes. However, whether the dynamic changes of ceRNAs can modulate miRNA activities remains controversial. Here, we examine the dynamics of ceRNAs during TGF-β-induced epithelial-to-mesenchymal transition (EMT). We observe that TGFBI, a transcript highly induced during EMT in A549 cells, acts as the ceRNA for miR-21 to modulate EMT. We further identify FN1 as the ceRNA for miR-200c in the canonical SNAI… Show more

“…FN1, derived from tumor-associated fibroblasts, stimulates cancer cell invasion after assembly [14]. In addition, as an intrinsic component of epithelialmesenchymal transition (EMT) regulatory circuit, FN1 is a potential target for interfering EMT during tumorigenesis 15 BioMed Research International [15]. Here, we suspect that FN1 is related to the pathway of ECM-receptor interaction.…”

“…We were able to distinguish low-risk and high-risk samples with a relatively high prognosis accuracy consisting of these six mRNAs. Among these mRNAs, TGFBI was investigated as a highly induced transcript during EMT in A549 cells, acting as the ceRNA for miR-21 to modulate EMT [36]. SFRP1 exhibited a tumor-promoting function by selectively activating TGFβ signaling in gastric cancer cells and thus activating EMT progress [37].…”

“…Epithelial-mesenchymal transition (EMT) plays an essential role in embryonic development and the transformation of early-stage tumors into invasive malignancies (4,5). Previous studies have demonstrated that TGF-β1 signaling is a potent inducer of EMT in various types of cancer, including NSCLC (6)(7)(8). It is well known that transforming growth factor β1 (TGF-β1) plays crucial roles in cell differentiation, proliferation, apoptosis and angiogenesis (9)(10)(11).…”
