Identification of Potential Biomarkers for Thyroid Cancer Using Bioinformatics Strategy: A Study Based on GEO Datasets

Abstract: Background. The molecular mechanisms and genetic markers of thyroid cancer are unclear. In this study, we used bioinformatics to screen for key genes and pathways associated with thyroid cancer development and to reveal its potential molecular mechanisms. Methods. The GSE3467, GSE3678, GSE33630, and GSE53157 expression profiles downloaded from the Gene Expression Omnibus database (GEO) contained a total of 164 tissue samples (64 normal thyroid tissue samples and 100 thyroid cancer samples). The four datasets w… Show more

“…GNAI1 belonged to the Gai family, which primarily functioned as inhibitors of adenylyl cyclase. GNAI1 was reported to confer hydrogen peroxide-induced apoptosis in human lung cancer cells and be associated with the prognosis of thyroid cancer patients (29,30). However, the biological functions of GNAI1 in GC remained unclear.…”

“…Using bioinformatics strategy, Shen et al found that fibronectin (FN1), neuromedin-U (NMU), chordin-like protein (CHRDL1), guanine nucleotide-binding protein subunit alpha-14 (GNA14), vasopressin V1a receptor (AVPR1A), guanine nucleotide-binding protein G(i) subunit alpha-1 (GNAI1), and integrin alpha-2 (ITGA2) may be potential biomarkers for the treatment of thyroid cancer. [ 16 ] Zou et al clarified that zinc finger protein 566 (ZNF566), zinc finger homeobox protein 3 (ZFHX3), PDZK1-interacting protein 1 (PDZK1IP1), and pituitary homeobox 2 (PITX2) are significantly associated with new biomarkers of atrial fibrillation-related stroke through the bioinformatics analysis of gene expression Omnibus (GEO) dataset. [ 17 ] Besides, Qian et al analyzed the microarray data of GSE59867 and found that proto-oncogene c-Fos (FOS), integrin alpha-IIb (ITGA2B), thrombospondin-1 (THBS1), and interleukin-8 (CXCL8) may play a vital role in development of heart failure after acute ST-segment elevation myocardial infarction.…”

“…The choice of therapy is a very crucial step after the identification of genes that play a critical role. Thus, we extracted drug molecules which could re-modulate the overexpressed and underexpressed genes, using the ‘Cmap2 database [45,46] as done in [47]. We queried a list of probe ids corresponding to upregulated genes (CLEC1B, CLEC3A, FCN1, NLRP10) and downregulated genes (RIPK2, SARM1, IRF7, CTSB, NLRP9) as input to Cmap2.…”
