Compatibility studies of hydrocortisone with excipients using thermogravimetric analysis supported by multivariate statistical analysis

Abstract: Application of thermogravimetry (TG) alone to study compatibility/incompatibility of active pharmaceutical ingredients (APIs) with excipients yields to misleading results due to overlapping of the thermal stages in the course of decomposition of both ingredients and their pharmaceutical mixtures. Hence, the purpose of this study was to assess the usefulness of multivariate statistical analysis as a supporting tool for interpretation of the TG traces during assessing compatibility of hydrocortisone as an API wi… Show more

“…It should be noted that in case of inert atmosphere, such products as NO, NO 2 , CH 3 COCH 3 could be obtained by intramolecular oxidation or some chemical groups rearrangements. In Table 4, fixed ions placed on the descending intensity values according to MS and well correlated with literature data [56][57][58][59][60][61][62][63][64][65][66][67].…”

“…This indicates that there is no interaction between the tested API and the excipient. Other authors recorded a similar relationship for diclofenac sodium [20], indapamide [21], enalapril [24], hydrocortisone [25] confirmed by the other analytical methods like DSC, XRDP, FTIR. The TG/DTG curves recorded for the tested mucolytic drugs, lactose monohydrate and physical binary mixture API with lactose monohydrate in different weight ratio are presented in Figure 2.…”

“…The IR spectrum of cortisol reveals the most pronounced absorption bands associated with valence vibrations of C=O groups [ 42 – 43 ]. According to [ 44 ] the calculated (experimental) frequency positions of the bands are located at 1686 (1660) cm −1 and 1725 (1735) cm −1 and demonstrate the highest intensities. Therefore, the structural parameters of the nanoantennas were chosen in such a way to ensure the LSPR absorption band close to the energy of the most pronounced vibrational modes in cortisol.…”
