Comparison of Two Wet Digestion Procedures for Multi‐element Analysis of Plant Samples

Abstract: Two microwave digestion systems for multi-element analysis of leaves and fruits were compared. Samples were digested by two different methods: 1) with HNO 3 (8 mL) and HClO 4 (6 mL) using an open microwave system and 2) with HNO 3 (5 mL) and H 2 O 2 (3 mL) in a closed microwave system. Total concentrations of aluminum (Al), cadmium (Cd), copper (Cu), chromium (Cr), iron (Fe), leaf (Pb), magnesium (Mg), manganese (Mn), nickel (Ni), and zinc (Zn) were determined in 22 samples by inductively coupled plasma atomic… Show more

“…Samples were analyzed with a Europa ANCA-GSL CN analyser (PDZ Europa Ltd, Sandbach, UK) following combustion at 1000 1C. Phosphorus concentration of finely ground oven-dried grain samples were measured using a Spectro-CCD (charge coupled device) ICP-AES unit (Spectro CIROS, Kleve, Germany) following a series of digestions with concentrated HNO 3 and H 2 O 2 at three temperatures: 120 1C for 4 h, 130 1C for 2 h and, finally, 145 1C until a white colored residue was obtained (Oliva et al, 2003).…”

“…At 30 and 60 days of experimental period, the plants were harvested and separated into aerial part and roots, washed with distillated water, and placed in paper bags in order to be dried in forced air stove at 65°C during 48 h. After drying, the plants were triturated and samples were weighed for foliar analyses. Plant samples were submitted to cooper determination conducted after nitric-perchloric digestion, according to Embrapa procedures (Embrapa, 1997), using a Inductively Coupled Plasma Optical Emission Spectroscopy (ICP OES), as described by Oliva et al (2003). The translocation index (TI) was determined by using the follow expression (Abicheque and Bohnen, 1998):…”

“…After drying, they were ground in a Willey type mill and packed in plastic bags. The wet digestion of plant samples was performed in open system, using adaptation of the method described by Oliva et al (2003). The concentrations of Cd, Cu, Fe, Mn, Pb and Zn in extracts of plant and soil were determined by inductively coupled plasma atomic emission spectroscopy (ICP-OES) (Miyazawa et al, 2009).…”
