Comparison of Two Protocols of Carbon Tetrachloride-Induced Cirrhosis in Rats – Improving Yield and Reproducibility

Abstract: Despite being a cardinal experimental model, the induction of cirrhosis in rats by repeated exposure to carbon tetrachloride (CCl4) has low reproducibility. Here, we compared two models of cirrhosis induced by orogastric administration of CCl4 once (CCl4-1xWk) or twice a week (CCl4-2xWk) for 12 weeks in male Sprague-Dawley rats. Control rats received water instead of CCl4. Both CCl4 protocols similarly attenuated body weight gain (p < 0.01 vs. Control). Although both CCl4 protocols increased hepatic fibrosis, … Show more

“…Furthermore, hepatotoxicity of CCL4 model in the present work was accompanied with inflammation and fibrosis. Fibrogenesis is known to be associated with the necrosis and inflammation of the liver (Fortea et al, 2018;Wahid et al, 2018). CCL4 is considered one of the most used hepatotoxins in the experimental study of liver diseases.…”

“…IDWE Ixeris denticulate water extract immune, common bile duct obstruction and multi-factor combination method are the commonly used methods for preparing animal models of liver cirrhosis [24,25]. Among them, CCl 4 as a classical hepatotoxic substance is widely used in the preparation of liver cirrhosis model, which commonly used 40-60% CCl 4 -mixed oil solution twice a week for about 10 weeks [26]. In our study, 40% CCl 4 oil solution was used for establishment of the liver cirrhosis model through repeated intraperitoneal injection in small dosage.…”

“…However, liver index showed no difference among three CCl 4 -treated groups. Although many studies demonstrated that administration of CCl 4 markedly impacted on the body weight and liver-to-body weight ratio, the different dosage of CCl 4 , age and body weight of the rats, the duration of treatment made the diversity of consequences (Fortea et al, 2018;Umezu, S. Yuasa, & Sudoh, 1985). In addition, previous studies have shown that CCl 4 administration leads to hepatic lipid accumulation and hepatomegaly (Lu, Weng, Chen, & Sheen, 2017).…”
