Comparison of Two Methods for the Detection of Urinary Iodine Used in Epidemiological Studies

Abstract: Two methods for the determination of iodine in urine or serum based on the Sandell-Kolthoff reaction were compared. The first was an autoanalyser method (AAII, Technicon) where the mineralisation takes place in a continuous flow manner. This procedure was used at the Department of Clinical Chemistry of the University Hospital of Berne (Switzerland) from 1968 until 1993. The second method was evaluated and adapted in our own laboratory. Each sample and the iodate-standards are mineralised in a Pyrex glass tube … Show more

“…UIC was determined by the Sandell±Kolthoff method adapted to modern laboratory equipment (21) by a well trained technical staff that had also performed the analyses of previous Swiss studies (1, 2, 13±16, 21,22). The coefficient of variation of the iodine assays was 7.5% (within-run precision).…”

“…Absorption was measured after a reaction time of 30 min by an automatic microwell-plate reader (SLT-Labinstruments spectrophotometer) at 405 nm. Three replicates of each sample were analyzed consecutively (21,22).…”

“…Automated modes of the Sandell-Kolthoff reaction for measurement of urinary iodine using Technicon AutoAnalyzer II (AAII) systems, with either dialysis or acid digestion, were compared with a method employing manual alkaline ashing. Another comparison of an autoanalyzer method (AAII), where the mineralization takes place in a continuous flow manner with a manual mineralization and discoloration in a 96-well microtiter plate read by a PC-controlled photometer, was performed by Wuethrich et al (2000). However, a comparison of five methods, with both manual and improved automatic digestion, as well as with both manual and automated reading of the Sandell-Kolthoff reaction, yielded agreement among the methods tested, and also with an inductively coupled plasma mass spectrometry method (May et al, 1997).…”

“…These can provide high sample throughput and reduce the amount of waste per sample [9][10][11], although the Technicon Autoanalyzer and equivalent segmented flow analyzers are still in use [7].…”
