Comparison of two means of attachment of an organometallic acid on gold surfaces by combining X-ray photoelectron spectroscopy and IR reflection spectroscopy

Abstract: International audienceTwo methods of immobilization of the butyric 4-oxocyclopentadienylmanganese tricarbonyl acid I on gold surface are presented: either through the assembly of a disulfide derived from the acid, or by coupling on a cystamine monolayer; the efficiency of these two ways are evaluated by IR reflection spectroscopy and X-ray photoelectron spectroscopy

“…In earlier works, cystamine and cysteamine were employed as bifunctional compounds, where, thanks to its high affinity for gold, the sulfur atom coordinated to the metal atoms at the surface while the amine function served as anchoring group to covalently attach other molecules to the thiolate surface (19,(53)(54)(55). This step constituted the starting point for the development of a number of biosensors (13,56).…”

“…This property was successfully exploited to develop enzymebased (9-13) and antibody-based (14-18) biosensors. We have already studied the direct chemisorption of manganese tricarbonyl disulfide compounds (19) and the binding of alkyne dicobalt hexacarbonyl labeled poly(L-lysine) to self-assembled monolayers of 11-mercaptoundecanoic acid on gold substrates (20). In both systems, transition metal carbonyl entities could be detected by FT-IRRAS and used as sensitive probes at gold surfaces.…”

“…The high-affinity biotin/avidin biorecognition system (K a = 1 × 10 15 M −1 ) is indeed a relevant ligand-receptor model because of its frequent use in immunoassays either for antigen/antibody immobilization or at the detection step (11). Moreover, transition metal carbonyl complexes give rise to intense vibration bands ν CO in the 1800-2150 cm −1 spectral range where few other molecules absorb, allowing their detection in solution (12)(13)(14)(15) and on a solid substrate down to the picomole level (16). Finally FT-IRRAS is a very sensitive and powerful technique for analyzing ultrathin organic films at the surface of reflective materials (17,18).…”

“…One-pot attachment onto the gold surface was used as a more suitable methodology for straight linkage of the cymantrene biomolecule conjugate via a cysteine terminated peptide (Scheme 1) instead of a two-step grafting approach via a functionalized thiolate intermediate. 45 Therefore, we have selected water and ethanol as solvents for solubility reasons and for further possible biological developments in the case of water.…”
