Comparison of Two Apheresis Systems of COBE and Optia for Autologous Peripheral Blood Stem Cell Collection

Abstract: Peripheral blood stem cell (PBSC) transplantation following myeloablative therapy is a mainstay of treatment for various types of malignancies. This study aimed to evaluate the differences between the Optia MNC and COBE Spectra MNC systems (Terumo BCT, Japan) according to apheresis procedures and the parameters of apheresis, products, and collection. The clinical data of 74 patients who underwent autologous PBSC collection from July 2012 to July 2015 were reviewed retrospectively. The patients comprised 48 (65… Show more

“…Our results are consistent with findings in the literature that have demonstrated equivalent therapeutic depletion results between the COBE Spectra and the Spectra Optia; Additionally, we found no significant difference in outcome parameters between the 2 apheresis systems. These findings are consistent with the published literature, which shows that hematocrit and platelet counts exhibit a periprocedural decline that is consistent across apheresis systems and leukapheresis protocols …”

“…Descriptive summary statistics included mean (standard deviation) and median (range) for continuous variables and number (percentage) for categorical variables. The threshold of <40% was determined a priori to be the clinically meaningful outcome of lymphocyte CE based on our institutional criteria for minimum acceptable operator and instrument performance and a conservative estimate based on the published literature …”

“…Analysis of the HPC product composition showed platelet contents were significantly lower in products collected from PM compared to GM group. Even though Optia system is more sparing of platelets, as previously reported, it appeared that the mobilization regimen has an impact on the product platelet content …”

“…Even though Optia system is more sparing of platelets, as previously reported, it appeared that the mobilization regimen has an impact on the product platelet content. 14,20 Our study is limited because it involved retrospective data collection and analysis, and therefore, we could not control for mobilization regimen and apheresis device selection. In addition, plerixafor was only added to the mobilization regimen when donors failed to be mobilized adequately with GCSF alone.…”
