Comparison of the proteomes of three yeast wild type strains: CEN.PK2, FY1679 and W303

Abstract: Yeast deletion strains created during gene function analysis projects very often show drastic phenotypic differences depending on the genetic background used. These results indicate the existence of important molecular differences between the CEN.PK2, FY1679 and W303 wild type strains. To characterise these differences we have compared the protein expression levels between CEN.PK2, FY1679 and W303 strains using twodimensional gel electrophoresis and identified selected proteins by mass spectrometric analysis. … Show more

“…Because we did not detect an accumulation of ERCs, we measured the average lifespan in our hho1 Δ cells to address the possibility that the shortened lifespan of hho1Δ mutants is a strain‐dependent phenotype. Different phenotypes in W303 and S288C strains with the same mutant allele have been observed previously (for example, see Rogowska‐Wrzesinska et al ., 2001). We analysed lifespan in wild‐type, hho1Δ and sir2Δ cells of the S288C genetic background (Fig.…”

“…A few other studies have shown that different S. cerevisiae genetic backgrounds result in significant physiological differences, e.g. in lipid metabolism (Daum et al 1999) or protein expression (Rogowska-Wrzesinska et al 2001). Another good example is the comparison of ENA/PMR2 loci encoding the alkali-metal cation ATPase (Wieland et al 1995;Daran-Lapujade et al 2003).…”

“…Variant D differs in a single nucleotide substitution from variant A and is present only in laboratory strains S288c and its derivative W303 (Rogowska-Wrzesinska et al 2001). S288c, the most popular genetic background in yeast research laboratories, is a derivative of a natural heterothallic diploid strain isolated from rotting figs in California in 1938 (Mortimer and Johnston 1986).…”
