Comparison of the Peroxidase‐Like Activity of Unmodified, Amino‐Modified, and Citrate‐Capped Gold Nanoparticles

Abstract: The origin of the peroxidase-like activity of gold nanoparticles and the impact of surface modification are studied. Furthermore, some influencing factors, such as fabrication process, redox property of the modifier, and charge property of the substrate, are investigated. Compared to amino-modified or citrate-capped gold nanoparticles, unmodified gold nanoparticles show significantly higher catalytic activity toward peroxidase substrates, that is, the superficial gold atoms are a contributing factor to the obs… Show more

“…Catalyzed reporter deposition techniques such as tyramide signal amplification (TSA) are commonly used to improve sensitivity of standard microarrays (Vora, Meador, Anderson, & Taitt, 2008). We have utilized TSA in our own laboratory on arrayed planar waveguides, with >100-fold improvement in observed detection limits (Figure 17.3); incorporation of stable nanoparticles exhibiting peroxidase activity would potentially accomplish similar tyramide deposition (Wang et al, 2012). Similar enzyme-based amplification schemes, such as PCR, rolling circle amplification (RCA; Nallur et al, 2001), and immunoRCA (Schweitzer et al, 2000), also show tremendous potential for improving sensitivity but to our knowledge have not yet been tested on TIRF-based planar waveguide platforms.…”

“…This phenomenon of auto-oxidation of gold and silver was previously established by different research groups. 29,30 Hence, we chose to use the aqueous gold colloid with an average particle size of 60 nm with the oxidized product TMB 2+ to enhance the Raman signal, and used the ultrasensitive SERS modality to quantify the peroxidase activity of Hb-Hp complex and thus the concentration of Hp. Figure 2A represents the SERS spectra of the substrate TMB and its oxidized version TMB stretching and CH bending vibrations, respectively.…”
