Comparison of the effects of epithelium removal and of an enkephalinase inhibitor on the neurokinin‐induced contractions of guinea‐pig isolated trachea

Abstract: 1 The influence of epithelium removal and/or thiorphan on the effects of neurokinins (substance P (SP), neurokinin A (NKA), neurokinin B (NKB)) and related peptides on airway contractility was investigated on the guinea-pig isolated trachea. 2 Removing the tracheal epithelium significantly enhanced the sensitivity but not the maximum contractile responses to the peptides.3 After removal of the epithelial layer, the shifts to the left of the log concentration response curves were greater for SP and SP-OMe (1.62… Show more

“…NKA may have been caused by a higher susceptibility of SP to degradation by peptidases, as reported in guinea-pig airways (Martling et al, 1987;Devillier et al, 1988). Our own results support this idea: the increases in potency produced by peptidase inhibitors were greater for SP (3.5 times) than for NKA (2.8 times).…”

“…Therefore, since no significant differences were observed between the 30 and 60min challenge period (P > 0.05), the 60 min period was selected for all subsequent experiments. The enkephalinase inhibitor, thiorphan has been shown to enhance the effector responses to exogenous and endogenous tachykinins in different muscle tissues (Devillier et al, 1988;Maggi et al, 1989). …”

“…Removal of epithelium from tracheal or bronchial strips potentiates the contractile response to SP and NKA by inhibiting tachykinin cleavage and inactivation (Devillier et al, 1988;Frossard et al, 1989). In addition, pretreatment of airway tissue with phosphoramidon, a potent inhibitor of NEP, significantly enhances tachykinin-induced contraction of the smooth muscle (Black et al, 1988;Lotvall et al, 1991).…”
