This work aimed to describe the activity of paraoxonase 1 (PON1) in serum, follicular fluid and seminal plasma of sheep. Average serum PON1 activity was 286.8 ± 96.2 U/ml in females and 237.6 ± 18.9 U/ml in males. There was a positive correlation between PON1 activity in serum and follicular fluid in females, being twice higher in serum than in follicular fluid (148.8 ± 15.7 U/ml). PON1 activity in males' serum was 10-fold higher than in seminal plasma (21.18 ± 14.2 U/ml), and there was no correlation between PON1 activity in both compartments. Finally, this work suggests that PON1 activity of in sheep is higher compared to other mammalian species, and there is an association between PON1 in serum and follicular fluid only.

Studies on Paraoxonase-1 Isolated from Amniotic Fluid and its Effect Against Cisplatin-Induced Hepatotoxicity and Cardiotoxicity in Rats

Paraoxonase-1 (PON1) was isolated from the amniotic fluid of normal pregnancies at last week of gestation. One proteinous band had been isolated by gel filtration sephadex)G-50) from the protein precipitate produced by ammonium sulfate saturation (67%) after dialysis. The product from (G-50) gave two bands by sephadex (G-100). It was found that the first peak (Peak A) had higher activity for (PON1). The apparent molecular weights of the isolated PON1 using gel filtration chromatography and SDS-PAGE was (43873 + 350) and (43682 + 278) Dalton respectively. The results also showed that the optimum conditions of PON1 was obtained at (80 µg/ml) of protein as a source of the enzyme using (10 mmol/l) of paraoxon as a substrate, Tris-HCl buffer (0.14 mol/l) as a buffer at pH (8.0) and incubation for (7) minutes at (45C). Using lineweaver-Burk plot, the values of maximum velocity (V max) and Michaelis constant (K m) were (80.0 µmol/ min) and (3.79 mmol/l) respectively. The protective effect of PON1 against cisplatin-induced hepatotoxicity and cardiotoxicity by intraperitoneal injection of (10mg/kg) cisplatin were evaluated in 35 male albino white rats classified into 5 groups. The rats were treated with 0.5mg/kg/day or 1mg/kg/day of isolated PON1 injected intraperitonealy for 5 successive days before and 5 successive days after induction of toxicity. The results showed a significant reduction in the levels of alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatine kinase (CK), total bilirubin(TB) and malondialdehyde(MDA) in comparison with the cisplatin treated animals. It was concluded that (PON1) protects the liver and heart against the toxicity induced by this cytotoxic drug.

Chronic Renal Failure (CRF) is the progressive and irreversible loss of normal functioning of kidneys. this is associated with increased levels of some biochemical parameters and decreased others. This study aims to evaluate the value of serum paraoxonase-1 (PON1) activity as biochemical marker for patients with chronic renal failure (CRF) before and after hemodialysis (HD). The samples of this study consists of 100 patients (55 males, 45 females) and 100 apparently healthy individuals (57 males, 43 females) from 20 - 70 years old of age. All patients included in this study were those who were receiving hemodialysis therapy in the dialysis unit in Kirkuk general Hospital from the period between February 2013 and May 2014. Serum PON1, BU, Cr, TC, TG, HDL, LDL levels were measured by spectrophotometric methods.Paraoxonase1 activity level was significantly lower in cases of CRF patients than in normal healthy control in pre hemodialysis (HD). It's level was significantly increased in post HD (P> 0.05). The blood urea (BU), serum creatinine (Cr), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) were significantly higher than normal healthy control in pre HD (P˂ 0.05), In post HD their levels were significantly decreased as compared to the pre HD (P˂ 0.05) but still lower than normal control, the serum high density lipoprotein (HDL) of CRF patients was significantly lower than normal controls (P˂ 0.05) in pre HD and was significantly increased in post HD as compared to pre HD but remained lower than normal healthy control. Conclusion: PON1 activity was significantly decreased in CRF patients before HD and significantly increased after HD but remained lower than controls. PON1 was significantly and negatively correlated with the age, BMI, duration of disease, BU, Cr, TC, TG, LDL, and VLDL. This study demonstrates that there is an increased risk of cardiovascular complications in CRF patients undergoing hemodialysis by increasing TC, TG, LDL, VLDL and decreasing HDL.
