Comparison of in vitro activity of danofloxacin, florlenicol, oxytetracycline, spectinomycin and tilmicosin against Mycoplasma mycoides subspecies mycoides small colony type

Abstract: Minimum inhibitory concentrations (MIC) and minimum mycoplasmacidal concentrations (MMC) of the antimicrobials danofloxacin, florfenicol, oxytetracycline, spectinomycin and tilmicosin were determined in vitro for 20 isolates of Mycoplasma mycoides subspecies mycoides small colony type (MmmSC), the causative agent of contagious bovine pleuropneumonia (CBPP). The majority of strains were most susceptible to tilmicosin, followed by danofloxacin, oxytetracycline, florfenicol and spectinomycin with MIC50 values of … Show more

“…A number of products have demonstrated efficacy against MmmSC in vitro, including oxytetracycline (OTC), danofloxacin and tilmicosin (Ayling et al, 2000(Ayling et al, , 2005. Modelling studies have predicted that antimicrobials, used in association with vaccination, could be successful in eradicating disease from infected herds (Mariner et al, 2006b).…”

“…The availability of recent MmmSC isolates was limited; however, 27 were isolated in Africa between 1996 and 2004. The remaining MmmSC isolates consisted of older strains and eight strains that had previously been tested against five of the antimicrobials used in this study (1). The field isolates had been minimally passaged in Eaton's broth medium (7) and stored at Ϫ70°C until required.…”

“…Species identification was confirmed by standard mycoplasma identification techniques (8) and by PCR (2). The MIC methods used were described previously (1). With the exception of tylosin (TYL), the antimicrobials were all obtained in pure form from Trek Diagnostic Systems (East Grinstead, West Sussex, United Kingdom) at specified concentrations on microtiter Sensititre plates.…”

“…Minimum inhibitory concentrations were initially determined for each drug against Mmm SC strain B237 to define a concentration range over which time-kill assays should be performed. To date, MICs have been published for only OTC and danofloxacin against a number of Mmm SC isolates, with MIC ranges of 0.125–4 mg/L and <0.06–0.5 mg/L for OTC and 0.125–1 mg/L and 0.12–0.5 mg/L for danofloxacin in artificial medium [11], [12]. Minimum inhibitory concentrations against Mmm SC have not been reported for tulathromycin, although Godinho et al .…”

“…Nevertheless, three major classes of antimicrobial are effective against mycoplasmas, namely tetracyclines, fluoroquinolones and macrolides [10]. Indeed, several antimicrobials have emerged on to the market since 1987, some of which show mycoplasmacidal activity in vitro [11], [12]. The in vivo efficacy of such antimicrobials against Mmm SC remains to be fully determined, as studies have been few.…”
