Comparison of HPLC and multivariate regression methods for hydrocortisone and lidocaine analysis of pharmaceutical preparations

Abstract: A reverse-phase high-performance liquid chromatographic (HPLC) method to determine hydrocortisone acetate, hydrocortisone hemisuccinate and lidocaine is described in this paper. The separation was made in a LichrCART C(18) column using a methanol-NaH(2)PO(4)/Na(2)HPO(4) (0.1 mol L(-1)) (pH=4.5) buffer solution as a mobile phase in isocratic mode (60:40 (v/v)). The mobile phase flow rate and the sample volume injected were 1 mL min(-1) and 20 micro L, respectively. The detection was made with a diode-array dete… Show more

“…The performance of various multivariate chemometric and derivative spectrophotometric methods is compared for examples of formulations containing hydrocortisone and ZnBacitracin 31 as well as dexamethasone and polymyxin B. 32 An interesting feature of another method for the assay of spironolactone and hydrochlorothiazide, based on ratio spectrum derivative spectroscopy, making use of a rapidscanning diode-array spectrophotometer, is that it is adopted to flow-injection analysis. 33 The importance of spectrophotometric/colorimetric methods based on chemical reactions has dramatically decreased in the last years both in the analysis of bulk steroid drugs and formulations made thereof.…”

“…However, modern chromatographic techniques such as high-performance liquid chromatography (HPLC) [812] and ultra-performance liquid chromatography (UPLC) [13] are preferentially employed for the determination of hydrocortisone in conventional pharmaceutical products such as tablets, creams, and injections. The different HPLC methods employ different chromatographic conditions including the use of complex and sometimes expensive solvent systems.…”

“…Out of various analytical methods, the multivariate regression spectrophotometry, micellar capillary electrophoresis (MEKC), and also RP-HPLC coupled with electrochemical detector to quantitation of these substances in combined dosage forms were applied. However, all of those studies have focused almost on the hydrocortisone acetate and lidocaine hydrochloride analysis in form of suppositories and ointments [15–17]. Therefore, attempts were made in this work to develop a fast, sensitive, robust, and cost-effective TLC-densitometric method suitable for quantitative analysis of hydrocortisone acetate and lidocaine hydrochloride in the commercial (injection) dosage form.…”
