Tropical Animal Health and Production,. 2 AbstractA cross-sectional study was performed in the livestock-wildlife interface areas of Lochinvar and Blue Lagoon National Parks and the non-interface area of Kazungula to determine the prevalence of antibodies to Brucella spp. in domestic ruminants and identify individual animal risk factors of infection. Atotal of 1245 cattle from 124 herds and 280 goats and sheep from 29 flocks were tested sequentially for Brucella antibodies using the Rose Bengal test (RBT) and competitive ELISA. In cattle, individual seroprevalence ranged from 14.1% to 28.1%, while herd sero-prevalence ranged from 46.2% to 74.0% in the three study areas. No goat or sheep tested positive for Brucella antibodies. Three types of cattle grazing strategies were encountered: locally grazed herds (LGH), transhumantly grazed herds (TGH) and river flood plain grazed herds (FGH). Brucella seroprevalence was seen to vary according to area and grazing strategy: Lochinvar and transhumant grazed herds recorded the highest figures, respectively. Age, sex and history of abortion were found to have independent effects on individual seroprevalence. This study establishes that brucellosis is endemic in domestic animals in the livestock-wildlife interface areas of Blue Lagoon and Lochinvar national parks and the disease is also present in Kazungula. We observed that type of grazing strategy had significant impact on cattle Brucella seroprevalence and that transhumant herds were at high risk of being infected.

Journal homepage: http://www.ijcmas.com Brucellosis is an important zoonotic disease globally that causes huge economic losses to the livestock owners and is of public health significance. Brucellosis in animals is endemic in India. In sheep and goats, Brucellosis is mainly caused by Brucella melitensis whereas Brucella ovis causes the disease in sheep. The symptoms in infected sheep and goats are abortions, stillbirths and the birth of weak offsprings. Animals that abort may retain the placenta. Sheep and goats usually abort only once, but reinvasion of the uterus and shedding of organisms can occur during subsequent pregnancies. Several studies have been carried out on seroepidemiology of caprine and ovine Brucellosis in India. The tests commonly used for diagnosis of Brucellosis are the milk ring test, Rose Bengal Plate Test (RBPT), Standard Tube Agglutination Test (STAT), Microtiter Plate Agglutination Test (MAT) and ELISA. The RBPT is a rapid screening test for the diagnosis of Brucellosis. The sensitivity of RBPT is very high (>99%) but the specificity can be low and it could sometimes give a false positive result. Its positive predictive value is low and a positive test result requires confirmation by a more specific test. One hundred and eight Nelore heifers (Bos indicus) about 18 months of age were vaccinated with a standard dosage (60-120 x 10 9 viable organisms) of Brucella abortus strain 19 vaccine, aiming at to study the persistence of serological antibody titres by the plate agglutination, rose Bengal and complement fixation tests. Serum samples were collected just before vaccination and after 45 days, 6, 12 and 18 months. Before the vaccination, all the heifers showed negative results by the three tests. After 45 days, all the heifers had titres higher than or equal to 1/100 by the plate agglutination test and all of them showed positive results by the rose Bengal plate test, while 2 animals had titre 1/2 and 106 animals had titres higher than or equal to 1/4 by the complement fixation test. One year after the vaccination, most of the heifers did not show significant serological antibody titres.Considering the risk to which animals in endemic areas and infected herds are subjected, and considering the clear reduction of the serological antibody titres shown by most of the vaccinated heifers, one may assume that, in case of Nelore heifers in endemic areas and that were not vaccinated between 3 and 8 months of age, it would be more reasonable to vaccinate them at 18 months of age than to expose them to a high risk of Brucella infection.INDEX TERMS: Bovine brucellosis, strain 19 vaccine, serological response.
