Comparison of Chromatin Assays for DNA Fragmentation Evaluation in Human Sperm

Abstract: Sperm chromatin integrity is vital for successful pregnancy and transmission of genetic material to the offspring. We evaluated chromatin integrity in sperm from 60 infertile men and 7 fertile donors comparing the sperm chromatin structure assay (SCSA), TdT-mediated-dUTP nick end labeling (TUNEL), the sperm chromatin dispersion (SCD) test, and acridine orange staining technique (AOT). The TUNEL and SCD assays showed a strong relationship with the SCSA (r Ͼ .866; P Ͻ .001) for sperm DNA fragmentation, both in i… Show more

“…We chose this commercial kit for the following reasons: the test was easy to set up and it did not require access to expensive equipment, which would have been unavailable in the routine laboratory where the study was carried out; this test also bears the CE marking. Moreover, it has been demonstrated that the sperm chromatin dispersion test is an effective tool for assessing sperm DNA fragmentation when compared with other tests [11,16,17]. The SCD test and TUNEL assay have the capacity to give similar results in terms of DNA fragmentation, and it has been demonstrated that the SCD test is even more sensitive than the TUNEL assay [18].…”

“…The Relationship between the Halosperm Assay and Semen Analysis Performed According to the 4 th and the 5 th Editions of the World Health Organization Guidelines men [10][11][12][13][14], poor fertilization rates [15,16], specific semen defects [17], miscarriage and poor artificial reproductive technology results [18]. Furthermore, infertile men have been shown to have substantially higher levels of DNA fragmentation than fertile men [13] and hence DNA fragmentation testing has been proposed as a valuable adjunct to routine semen analysis when considering the fertility potential of a man [16].…”
