Comparison of broadband UVB, narrowband UVB, broadband UVA and UVA1 on activation of apoptotic pathways in human peripheral blood mononuclear cells

Abstract: The greater effectiveness of UVB on initiation of apoptotic pathway suggests that apoptosis may play a role in the clinical efficacy of UVB-responsive inflammatory cutaneous diseases.

“…As previously shown [31], cell damages from UV light exposure strictly depends on source intensity and emitting spectrum. While short UV wavelengths (UVB range) consistently damage cells even at low energy levels (in the order of mJ/cm 2 ), the shifting to higher wavelengths dramatically reduces the incidence of cell death.…”

“…Irgacure 2959 is currently the most commonly used PI molecule for the generation of viable cell-laden constructs, due to its low level of cytotoxicity [27][28][29]. However, Irgacure 2959 presents an absorption peak in a wavelength range between 250 and 300 nm [30], which is known to induce significant cell mortality even at relatively low level of absorbed energy [31]. The search for photoinitiators minimizing ultraviolet (UV) light exposure as well as molecule cytotoxicity led to investigate alternative solutions [32].…”

“…It is possible that the higher doses needed in using NB-UVB phototherapy resulted in burning and cutaneous sensitivity of the patients, who subsequently did not experience irritation with the lower doses used for BB-UVB phototherapy. Both NB-UVB and BB-UVB induce apoptosis in a dosedependent manner but sources emitting a narrow band of wavelengths require a higher dose or fluence to induce apoptosis (32).…”

“…Both UVA and UVB share immunological effects that eventually result in vitiligo improvement. UVA1, which constitutes 80% of BB‐UVA , Waldmann H. GmbH & Co., unpublished data), suppresses tumor necrosis factor alpha and interleukin‐12 (IL‐12). Moreover, it decreases the levels of interferon gamma and intercellular adhesion molecule type‐1 which plays a major role in lymphocyte activation and trafficking .…”
