Comparison of antiinflammatory and antiallergic drugs in the melittin- and D49 PLA2-induced mouse paw edema models

Abstract: Melittin (MLT) (10 micrograms/paw) and D49 (0.4 micrograms/paw) were injected into the hind paw of male CD-1 mice and elicited 70-80% of maximal paw edema responses at 60 and 30 min after injection, respectively. D49 paw edema was significantly inhibited by anti-histamine/serotonin agents, a PAF antagonist, a PLA2 inhibitor, and some but not all 5-LO and CO inhibitors, indicating that this edema is produced by several classes of inflammatory mediators with mast cell degranulation apparently playing a major rol… Show more

“…On the other hand, direct injection of BV or its constituents has been shown to produce an inflammatory response. In this regard, intraplantar injection of melittin (10 µg/paw) in normal mice elicits 70-80% of maximal paw edema responses at 60 min after injection [14]. Thus based on this and other data cited above one can assume that BV might produce pain and inflammation under normal conditions.…”

“…PLA 2 (30 µg/paw), a major inflammatory component of BV (12% of dry wt), transiently evokes paw edema after subplantar injection [14,22]. As indicated above melittin has also been reported to induce paw edema after plantar injection in mice [22].…”

“…Although BV acupuncture therapy is relevant to treating various chronic pain diseases in Korean traditional medicine, the treatment sometimes results in erythema and eventually edema and pain, because of allergenic components in BV. It has been reported that melittin injection induces paw edema in mice (Hartman et al, 1991). Thus, the use of BV acupuncture therapy in treating disease has been somewhat limited.…”
