Lactoferrin (LF) is a major natural antimicrobial agent secreted in body fluids as a natural innate immunity protein. The action and structure of LF are closely related to its iron-binding capacity with structural reporting in open and closed conformations. This study looked at how lactoferrin structures change in camel (cLF), bovine (bLF), and human (hLF) lactoferrin closed forms after iron is removed from their binding sites. Initially, the sequence comparison between cLF and the LFs of marine mammals, bats, and domestic animals was the most intriguing conclusion. Camel LF is revealed to be more closely related to marine animals (~80.36% identity) and bats (~79.3% identity) than to terrestrial mammal species (~75.5% identity). Results indicated that cLF was more dynamic in nature than bLF and hLF by showing higher RMSD values. The cLF is known to be half lactoferrin half transferrin; in this study, we show that there are different MD behavior of both iron-binding sites. While LF contains two lobes (C- and N-lobes), the C-lobe showed high fluctuations as N-lobe was more stable in the absence of ferric ions. The C-lobe and N-lobe of cLF react differently at physiological pH, revealing distinct molecular interactions between these components. In addition, cLF showed higher system flexibility derived from its larger RMSD, RMSF, lower intermolecular hydrogen bonds, and higher solvent accessible surface area (SASA).

In addition to their nutritional properties, mushrooms have emerged as a health supplement because of their medicinal potential. Many studies have shown that mushrooms exhibit important biological activities. Here, the antioxidant and antimicrobial activities of Hohenbuehelia myxotricha (Lév.) Singer mycelia cultivated on Sabouraud dextrose broth (SDB) and glucose peptone yeast (GPY) medium were studied. The total antioxidant status (TAS), total oxidant status (TOS), and oxidative stress index (OSI) of ethanolic extracts of mycelia were measured using Rel Assay kits. The antioxidant and oxidant potentials of H. myxotricha mycelial extracts were determined for the first time in the present study. The highest TAS, TOS, and OSI values of H. myxotricha were 5.416 ± 0.150 mmol/l, 1.320 ± 0.156 μmol/l, and 0.024 ± 0.003, respectively. Ethanolic mycelial extracts of H. myxotricha showed antimicrobial activities at concentrations from 25 to 200 μg/ml against all the studied bacteria ( Acinetobacter baumannii, Enterococcus faecalis , Escherichia coli , Staphylococcus aureus , methicillin-resistant S. aureus , and Pseudomonas aeruginosa ) and fungi ( Candida albicans , C. glabrata , and Issatchenkia orientalis ) tested by the agar dilution method. The antifungal activity of the extract was more significant than its antibacterial activity. The antioxidant, oxidant, and antimicrobial potentials of H. myxotricha mycelia varied depending on the culture media used. GPY medium was more suitable for the synthesis of antibiotic compounds against E. coli , while SDB medium was more appropriate for producing metabolites with antioxidant and antifungal properties. Based on the results, ethanolic extract of H. myxotricha mycelia showed a significant pharmacological potential and could be used as a natural antioxidative and antimicrobial source for health benefit.

Plant extracts and essential oils extracted from several medicinal plants like Lippia citriodora, Bunium persicum, Thymus vulgaris, and Mentha piperita were successful in preventing mycelial growths of Trichoderma harzianum under both in vitro conditions (Tanhaeian et al, 2020) and in vivo conditions in the Agaricus bisporus and P. ostreatus farms (Hatvani et al, 2012). Mushrooms are known for several nutritional and medicinal benefits and are cultivated worldwide. Several fungal contaminants of mushrooms have been serving as the major restraining factor in the growing mushroom industry for a long time. Fungal contaminants like Trichoderma spp., Mycogone spp., Lecanicillium spp., Cladobotryum spp., Coprinus spp., Sependonium spp., Sclerotium rolfsii, and Cephalothecum roseum among many, are found to infect mushroom crops at different stages from spawn run period to maturation of fruiting bodies. These contaminants may reduce yield and/or degrade the quality of fruiting bodies of the mushroom causing economic losses. These contaminants are usually peculiar in terms of their symptomatology on the substrates, disease cycle, epidemiological requirements, and yield losses. Most of these contaminants come from poorly sterilized substrates. Several sterilization techniques like steam sterilization, hot water sterilization, alkalinization, bleaching, and chemical sterilization can be employed to eliminate pre-existing contaminants and each technique has its own relative advantage over others. Besides, biological control involving botanicals and live antagonists can also be used as prophylactic sterilant or as therapeutic sprays. Biological control measures are friendly to the environment and human health. Unlike chemical fungicides (used as sterilant or spray), biological control measures don’t inhibit mushroom mycelial growth and even don’t raise the problem of pesticide resistance in pathogens. Roguing out of infected mushroom fruiting bodies or beds, mushroom house sanitation, and management of vector population are also equally important in preventing the spread of the fungal diseases of mushrooms.
