Comparative studies on the fermentative production of lantibiotics by staphylococci

Abstract: The production of the lanthionine-containing polypeptide antibiotics gallidermin from Staphylococcus gallinarum TU 3928 and pep 5 from S. epidermidis 5 is investigated with respect to regulation and stimulation of productivity by media components, optimization of both the media used and the fermentation process and is compared to the production of the lantibiotic epidermin from S. epidermidis TU 3298. Efficient methods for rapid quantification of lantibiotics, optimization of the media and a primary enrichment… Show more

“…All these characteristics of nisin production clearly differentiate it from that of authentic secondary metabolites (Kleinkauf et al, 1986;Martin & Liras, 1989). Almost all bacteriocins produced by lactic acid bacteria are produced during the active growth phase; further, the lantibiotics Pep 5 (Sahl & Brandis,198 l), ancovenin (Kido et al, 1983), epidermin (Horner et al, 1989) and gallidermin (Horner et al, 1990) are also synthesized during the growth phase of the producer strain.…”

“…The time indications (l), (2) and (3) correspond to the arrows in (a). The blot was hybridized to the radioactively labelled natural ancovenin (Kido et al, 1983), epidermin (Horner et al, 1989) and gallidermin (Horner et al, 1990) have been shown to decrease as soon as the maximal antibiotic titres were reached. The decrease of the nisin titre after 8 h of fermentation implies that the fermentation process would need to be stopped by then to achieve a maximum specific nisin productivity.…”

“…Gallidermin is a tetracyclic, 21-amino-acid, type A lantibiotic peptide which is produced by the nonpathogenic Staphylococcus gallinarum Tü3928 (25) and shows broad-spectrum activity against important gram-positive pathogens like S. aureus, Staphylococcus epidermidis, Streptococcus pyogenes, Streptococcus pneumoniae, Streptococcus faecalis, and P. acnes (20). Gallidermin (6L-epidermin) is closely related to epidermin, which is synthesized by the class II strain S. epidermidis Tü3298 (1,51).…”
