Comparative proteomic analysis of antimony-resistant and -susceptible Leishmania braziliensis and Leishmania infantum chagasi lines

Abstract: The emergence of drug-resistant Leishmania species is a significant problem in several countries. A comparative proteomic analysis of antimony-susceptible and antimony-resistant Leishmania braziliensis (LbSbR) and Leishmania infantum chagasi (LcSbR) lines was carried out using two-dimensional gel electrophoresis (2-DE) followed by mass spectrometry (LC/MS/MS) for protein identification. Out of 132 protein spots exclusive or up-regulated submitted to MS, we identified 80 proteins that corresponded to 57 distinc… Show more

“…Several genes have been described to be involved in virulence and resistance to therapy (22,(25)(26)(27). Therefore, some of those genes related to treatment resistance and parasite virulence were also assessed.…”

“…Therefore, some of those genes related to treatment resistance and parasite virulence were also assessed. On the one hand, alpha-tubulin, which had been demonstrated to be more abundant in Sb(III)-resistant Leishmania strains (27), was analyzed. Interestingly, its gene expression was significantly reduced after treatment.…”

“…The proteomics analysis in Leishmania spp. showed that its mechanism of antimony resistance is complex, incorporating aspects of protein folding/chaperones, stress response, antioxidant/detoxification, diverse metabolic processes, RNA/DNA processing, and de novo protein biosynthesis (135)(136)(137)(138). The proteomic study of A. tumefaciens GW4 revealed that Sb(III) could influence the Ars resistance, the Sb(III) oxidase AnoA, phosphate metabolism, carbohydrate transport and metabolism, and the metabolism of lipids, purines, and amino acids ( Table 1).…”
