CRP (Citri Reticulatae Pericarpium), a famous traditional Chinese medicine, has also been extensively used in foods and condiments in dietary practice for centuries. According to the Chinese Pharmacopeia (2015 edition) it contains two subtypes, Guangchenpi (GCP) and Chenpi (CP). GCP exclusively originates from the pericarp of Citrus reticulata 'Chachi' cultivar and it's generally believed that GCP has superior qualities compared with the other main cultivars (CP). In the present study, an integrated approach combining LC-QTOF MS-based untargeted metabolomics analysis and DNA barcoding molecular identification was conducted to study the genetic diversity and chemical differences between GCP and CP. A validated UPLC-QTOF MS metabolomics method was established to identify markers by using PCA and OPLS-DA models. 34 identified metabolites could be used as chemical markers to distinguish effectively between the two subtypes. Among them polymethoxyflavones (PMF) such as hexamethoxyflavone (nobiletin and natsudaidain), pentamethoxyflavone (tangeretin and sinensetin), and tetramethoxyflavone are the most influential markers. Support vector machines were employed to classify all the samples and these markers showed good prediction accuracy (100%). The results of DNA barcoding showed that the secondary structure of the ITS2 sequences were significantly different among GCP and other three cultivars. The study indicated the integrated method could be a powerful and reliable analytical tool for differentiating GCP from CP.

Background Intrauterine adhesion (IUA) is a consequence of endometrium damage, leading to infertility, recurrent miscarriage, and various complications of pregnancy. IUA usually occurs after an infection or injury-related inflammation of the endometrium. CXCL5, a member of chemokines, is associated with the remodeling of connective tissues, but its roles in the formation of IUA remain uncertain. This study aims to investigate the expression and effect of CXCL5 in the development of IUA. Results CXCL5 protein level in the endometrium of IUA rat is significantly decreased compared with that in normal and sham-operated endometria of rats (P < 0.001). Furthermore, real-time PCR and western blotting assays also show that the CXCL5 expression in endometrial tissues from IUA patients is much lower than that in normal endometrium (P < 0.01), which is consistent with the results based on the rat model. CXCL5 significantly up-regulates MMP9 expression and slightly up-regulates p65 expression in human endometrial cells after CXCL5 overexpression. Conclusions These results show that CXCL5 plays an important role in the inhibition of IUA formation after injury by modulating MMP9 expression and therefore has a protective effect of CXCL5 in the adhesion formation of IUA. These findings provide valuable information for evaluating prognosis and guiding therapy of intrauterine adhesions.

Migraine is a chronic brain disease that leads to periodic neurological attacks. Parishin A and isorhynchophylline (PI) is the active monomer component extracted from the traditional antimigraine Chinese medicinal combination of Gastrodia and Uncaria, respectively. In this study, using high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS) technology, we performed pharmacokinetic and lipidomic study on migraine model rats after administration of PI. For the detection of the compounds in plasma, AB Sciex Triple Quad™ 4500 was applied for quantitative analysis, and the COSMOSIL C18 column (2.1 × 100 mm, 2.6 μm) was used for separation. Isorhynchophylline (ISO: m/z 384.8–241.2) and its main metabolite rhynchophylline (RHY: m/z 384.8–160.2) were simultaneously detected under positive ion modes. Besides, parishin A (PA: m/z 995.1–726.9) and its main metabolite gastrodin (GAS: m/z 331.1–123.0) were simultaneously detected with negative ion modes. For the analysis of endogenous lipid components, Dionex Ultimate 3000 (UHPLC) Thermo Orbitrap Elite was applied for the detection, and the Waters UPLCRBEH C18 column (1.7 μm 100 ∗ 2.1 mm) was used for separation. Chloroform/methanol (2 : 1, v : v) was used for extraction. The results demonstrated that PI exists significant difference in metabolism between single- and coadministration and can regulate lipid levels associated with migraine.
