Radiological features and biochemical changes were investigated during the parenchymal and ductal phases of chronic Fasciola hepatica infection in sheep. The activities of plasma aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and gamma-glutamyl transpeptidase (GGT), plasma levels of IgG anti- F. hepatica and serial ultrasound, computed tomography (CT) and magnetic resonance imaging (MRI) findings were studied in eight sheep infected with 150 F. hepatica metacercariae. Experimental fluke infection provoked an increase in plasma level of IgG directed against F. hepatica and in plasma LDH and AST activities from 4 weeks after infection. Enzyme activities did not significantly differ from the baseline after 15 and 12 weeks for LDH and AST, respectively. GGT activity increased from 9 weeks postinfection and still remained significantly elevated at 18 weeks. In the parenchymal phase, both CT and MRI showed nodular lesions in five animals and MRI could also detect early tracks in the subcapsular area in three sheep. Ultrasound findings were nonspecific in this phase. Ductal dilatation was shown by ultrasound, CT and MRI in almost all animals, although MRI was inferior to CT in depicting a mild ductal dilatation. Moving echogenic forms in the dilated bile ducts were observed by ultrasound from 9 weeks postinfection in seven of the eight sheep. Moving worms were only demonstrated in four sheep at CT and in a single sheep at MRI. This study shows that radiological findings may be useful in studies of fluke-induced liver damage.

The activities of the drug metabolizing enzymes aminopyrine-N-demethylase, aniline hydroxylase and UDP-glucuronyl transferase, together with protein concentration, were measured in liver microsomes and duodenal mucosa from healthy chickens and chickens experimentally infected with Escherichia coli endotoxin, or naturally infected with Eimeria necatrix and Eimeria tenella (clinically classified as slight, moderate or severe infections). E. coli (2 mg/kg, 3 days) and severe coccidiosis significantly decreased the activities of the three enzymes in the liver and duodenum. However, infection by the E. coli endotoxin at lower doses (0.5 and 1 mg/kg, 3 days) and moderate or slight coccidial infections had no significant effect on the activities of these enzymes. Neither E. coli nor coccidial infections significantly affected the liver/body weight ratio. However, infection with the E. coli endotoxin (2 mg/kg, 3 days) and the moderate or severe coccidial infections significantly reduced microsomal protein concentration in the liver.

Ten female Churra breed sheep were experimentally infected with 25 Fasciola hepatica metacercariae daily during six days. The animals were offered pelleted lucerne and water ad libitum. Feed and water intake was automatically recorded by computer monitoring of electronic balances. Liver damage caused by flukes was ascertained by measuring the activities of aspartate amino transferase, glutamate dehydrogenase and gamma-glutamyl transferase. Serum bile acids and glucose concentrations were also measured. Maximum plasma enzyme values were found at 10 weeks post infection. Plasma bile acid concentration increased from six to 13 weeks post infection. Glycaemia was decreased from the fourth week post infection until the end of the experiment. Total daily food intake decreased maximally between six and 12 weeks post infection. The number of meals per day and time spent eating and drinking decreased along the experimental infection. The rate of eating and drinking increased progressively. An association between anorexia and liver damage is suggested.
