Comparative genetic diversity studies of Theobroma cacao L. using RFLP and RAPD markers

Abstract: The genetic diversity of 106 genotypes of Theobroma cacao L. was assessed using RFLP and RAPD methods. Thirty-one cDNA probes and 19 primers generated, respectively, 87 and 49poly-morphic bands that were submitted to factorial and cluster analyses. Both RFLP and RAPD bands separated the individuals into three distinctive groups. Some genotypes did not correspond to the currently accepted classification and are considered to have been originally misclassified. The nature of RAPD bands was investigated by Southe… Show more

“…This class also included material from germplasm collections in Costa Rica, Cô te d'Ivoire, Mexico, Venezuela and France. 'Modern Criollo' represents the genotypes studied as Criollo in previous biochemical and molecular studies (Lanaud, 1987;Bekele and Bekele, 1998;Ronning and Schnell, 1994;Laurent et al, 1995;N'Goran et al, 1994;De la Cruz et al, 1995;Lerceteau et al, 1997;Whitkus et al, 1998).…”

“…Therefore, markers provide an efficient method for genetic resources characterization, through which genetic diversity and organization at different levels can be assessed (Karp & Edwards, 1997;Lanaud & Lebot, 1997). RFLP has been broadly and successfully used to characterize genetic resources including some tropical perennial crops (Besse et al, 1994;N'goran et al, 1994;Jack et al, 1995;Lebrun et al, 1995;Luo & Boutry, 1995;Lerceteau et al, 1997). However, this technique requires a relatively large quantity of good quality DNA and the best results are obtained using radioisotopes, which makes RFLP technically and ecologically sensitive.…”

“…In general, accessions from the Upper Amazon germplasm show higher GD than those from other regions (MARITA et al, 2001;RONNING et al, 1995;N'GORAN et al, 1994;LERCETEAU et al, 1997;WHITKUS et al, 1998). The clone set studied here consisted of Upper and Lower Amazon and Trinitario germplasm, comprising 20 TSA-644 descendants and 10 non-descendants.…”
