Comparative Cytotoxicity of Glycyrrhiza glabra Roots from Different Geographical Origins Against Immortal Human Keratinocyte (HaCaT), Lung Adenocarcinoma (A549) and Liver Carcinoma (HepG2) Cells

Abstract: of incubation at 37C, the medium was discarded and replaced with 100 L of DMSO.The OD 570 was determined with a microplate reader (CLARIO Star Microplate reader, BMG Labtech, UK) and each experiment was repeated three times with five replicates in each experiment. The OD 570 obtained for the controls cells (no extract) was arbitrarily set at 100% and the OD 570 value for cells treated with extracts were expressed as a percentage of this control. The IC 50 values were calculated using

“…Cell lines, cell cultures and the 3-(4,5-dimethylthiazol-2-yl)-2,5-dphenyltetrazolium bromide assay. The potential cytotoxicity of C. reticulata stem bark extracts, their VLC fractions and the isolated compounds were studied against cancer cell lines A549 (human lung adenocarcinoma), MCF7 (human breast adenocarcinoma), PC3 (human prostate adenocarcinoma) and the normal cell line PNT2 (human prostate) using the MTT assay (Mosmann, 1983;Basar, Oridupa, Ritchie, et al, 2015). All cell lines were cultured in RPMI-1640 medium supplemented with 10% foetal bovine serum and 1% antibiotic-antimycotic solution (100 ×).…”

“…Briefly, RD cells suspension (100 µL) were seeded into 96-well plates and maintained in the incubator for 24 h. Subsequently, the cells, which obtained a confluency of about 90%, were treated with serial dilutions (100 µL each) of the mushroom extract, ranging from 1000 -0.01 µg/mL. The extract itself was previously dissolved in DMSO and diluted with the culture medium, ensuring the maximum concentration of DMSO was not more than 0.05%, a concentration that is not toxic to cancer cells 30 . The treatment was carried out for a period of 72 h in the same environment mentioned above.…”

“…Researchers have evaluated the cytotoxicity of polyphenol molecule extracted from Glycyrrhiza glabra which is used for the treatment of breast and prostate cancer, head and neck cancer cell lines and non‐malignant primary mucosal cells, and against different cancer cell lines such as Immortal Human Keratinocyte (HaCaT), Lung Adenocarcinoma (A549), and Liver Carcinoma (HepG2) cell lines …”
