Comparative analysis of virulence determinants and mass spectral profiles of Finnish and Lithuanian endodontic Enterococcus faecalis isolates

Abstract: A high prevalence of virulence factors was demonstrated in both population types. SELDI-TOF-MS and MALDI-TOF-MS proved useful in distinguishing between different E. faecalis phenotypes and they may be useful technologies for elucidating the eco-distribution of E. faecalis in humans.

“…Dentin Infection with Enterococcus faecalis E. faecalis VP3-181 isolated from a case of persistent apical periodontitis (19) was used as a test organism and grown in air at 37 C overnight on brain heart infusion agar (Becton-Dickinson, Sparks, MD) plates for the experiments. The bacteria were suspended in brain heart infusion (BHI) (Becton-Dickinson) broth and standardized spectrophotometrically to 3 Â 10 6 colony-forming units (CFU)/mL (OD 405 = 0.05).…”

“…Based on the spectral signatures of their surface components, a collection of 58 isolates was grouped by geographical location. Evidence of quinupristin-dalfopristin (Synercid) susceptibility was also provided (122). A second study utilized MALDI-TOF MS as a bacterial source-tracking tool to characterize environmental isolates of Enterococcus spp.…”

“…MALDI-TOF MS has been used for the characterization of enterococcal isolates from both human and environmental sources (122,123). Böhme et al documented the accuracy of the Bruker BioTyper system for identification of 30 Enterococcus spp.…”

“…Enterococcus faecalis can be recovered from the root canals of teeth with primary and secondary root canal infections (13,14). Clinical strains demonstrate various virulence characteristics (15,16), for example, gelatinase activity that might contribute to long-term survival of E. faecalis in obturated root canals (17). The species is commonly used to evaluate the antimicrobial efficacy of materials used in endodontic treatment, but frequently by using only a single strain from a nonendodontic source as opposed to multiple strains isolated from infected root canals.…”
