Comparative Analysis of Pdf-Mediated Circadian Behaviors BetweenDrosophila melanogasterandD. virilis

Abstract: A group of small ventrolateral neurons (s-LN v 's) are the principal pacemaker for circadian locomotor rhythmicity of Drosophila melanogaster, and the pigment-dispersing factor (Pdf) neuropeptide plays an essential role as a clock messenger within these neurons. In our comparative studies on Pdf-associated circadian rhythms, we found that daily locomotor activity patterns of D. virilis were significantly different from those of D. melanogaster. Activities of D. virilis adults were mainly restricted to the phot… Show more

“…This construct was expressed in a somewhat broader set of circadian neurons with the dvpdfgal4 driver; i.e., all PDF cells, LNds, and the PDF-negative fifth s-LNv (Bahn et al 2009). These flies also exhibit long periods, 1.3-h longer than control flies (Supplemental Fig.…”

“…All gal4 and gal80ts drivers used in this study have been described previously: timgal4 , pdfgal4 (Stoleru et al 2004), dvpdfgal4 (Bahn et al 2009), tubgal80ts (McGuire et al 2003, usp8-RNAi R2 (Mukai et al 2010), and usp8-RNAi (HMS01889, Bloomington). uas-usp8-DN transgenic flies were generated by injecting yw embryos with pUAST-usp8-DN plasmid (BestGene, Inc.).…”

“…Normal flies exhibit two daily peaks of activity: a morning peak that corresponds to lights on and an evening peak around the time of lights off. After entrainment, these patterns are retained under DD conditions (Hamblen-Coyle et al 1992;Renn et al 1999;Bahn et al 2009), providing a 23-to 24-hr circadian cycle period. Results obtained for a variety of genotypes are presented in Table 1.…”

“…The number of PDF--positive neurons, including both I--LNvs and s--LNvs neuronal groups in the brain, and their projection patterns appeared normal in the two mutant strains. Whole--mount anti--PDF--immunohistochemistry was performed in dissected adult brains, as described in BAHN et al 2009. A) wildtype (isoCJ1 strain) animals (10 brains examined); B) CRMP supK1 animals (11 brains examined); C) CRMP supIa1 animals (10 brains examined).…”

“…Several other insect species have been 72 found to show unimodal activity pattern, and thus their seasonal time measuring cannot be based 73 on above-mentioned system. For example housefly Musca domestica (Helfrich et al, 1985) and 74 some D. virilis group species (Bahn et al, 2009;Kauranen et al, 2012) show only evening 75 activity peak, while D. ananassae shows only morning activity peak (Joshi, 1999). 76…”
