Combined Effects of Soy Isoflavones and β-Carotene on Osteoblast Differentiation

Abstract: Soy isoflavones, genistein, daidzein and its metabolite equol, as well as β-carotene have been reported to be effective for maintaining bone health. However, it remains to be elucidated whether combining soy isoflavones with β-carotene is beneficial to bone formation. This study investigated the combined effect of soy isoflavones and β-carotene on the differentiation of MC3T3-E1 preosteoblastic cells. Daidzein and genistein alone did not affect cell growth but increased alkaline phosphatase (ALP) activity. Bet… Show more

“…26,27 The ability of b-carotene to induce osteogenic differentiation of stem cells has been reported. [28][29][30] Although there are some recent articles about encapsulation of b-carotene within electrospun bers, [31][32][33] based on our literature survey, no research has been reported on the incorporation of b-carotene within electrospun scaffolds for tissue engineering applications.…”

“…RARs attach to DNA as heterodimers with the retinoid X receptors (RXRs) and consequently modulate retinoic acid-responsive target genes. 28,[39][40][41] Wang et al 2 reported that b-carotene exerted an inhibitory effect on osteoclastogenesis and resorption pit formation through the nuclear factor-kappa B (NF-kB) pathway. To study the osteoinductive effect of b-carotene-containing mats on the differentiation of MSCs, scaffolds (PCL/bC2% and PCL/bC4%, 0.5 Â 0.5 cm 2 ) were seeded with cells (10 4 cell per well) and kept in DMEM with 10% FBS as a non-differential medium for up to 21 days.…”

“…The ingredients of propolis that are responsible for elevated level of the ALP enzyme are isoflavonoids, such as daidzein and genistein. Isoflavonoids are natural endocrine active compounds generally considered to promote health, and prevent or slow the onset of certain chronic diseases, such as osteoporosis (NISCHIDE et al, 2015;ZHENG et al, 2017). A number of studies have reported that soy isoflavonoid dose-dependently inhibits bone loss in both female and male osteoporotic animal models (REINWALD and WEAVER, 2006).…”
