Dental caries is the most prevalent human infection. It is a multifactorial disease in which the microbial composition of dental plaque plays a major role in the development of clinical symptoms. The bacteria most often implicated in the development of caries are that group of streptococci referred to as the mutans streptococci, in particular Streptococcus mutans and Streptococcus sobrinus. One approach to the prevention of caries is to reduce the numbers of mutans streptococci in plaque to a level insufficient to support demineralization of the tooth. In this study, zoocin A, a peptidoglycan hydrolase, combined with lauricidin, a cell membrane active lipid, was shown over a 72 h period to selectively suppress the growth of S. mutans in a triple species biofilm. Growth of the non-target species Streptococcus oralis and Actinomyces viscosus was not inhibited. In treated systems the amount of extracellular polysaccharide matrix produced was much reduced as determined by use of fluorescein isothiocyanate conjugated wheat germ agglutinin. The pH of treated biofilms remained above neutral as opposed to a value of 4.3 in untreated controls. We conclude that use of antimicrobial compounds that specifically target cariogenic bacteria should be further explored.

Background: Dental caries is one of several prevalent oral diseases caused by dental plaque biofilms. This study evaluated the anti-cariogenic effects of a bamboo salt (BS) and sodium fluoride (NaF) mixture on oral bacteria. Methods: The effects of several mixtures of NaF and BS on acid production, growth, and adhesion to glass beads of Streptococcus mutans, and their anti-cariogenic properties were investigated. The growth of S. mutans was measured according to optical density at 3, 6, 9, 12, 15, 18, and 24 hours after treatment using spectrophotometry at a wavelength of 600 nm, while pH was measured using a pH meter. Adhesion of S. mutans was measured according to the weight of glass beads from each group before and after incubation. Gene expression was measured using real-time polymerase chain reaction. Acid production and growth patterns of S. mutans were compared using repeated measures analysis of variance, followed by Scheffe's post-hoc test. The Kruskal-Wallis test was used to compare adhesion, followed by the Mann-Whitney test. Gene expression in the experimental and control samples was compared using the Student's t-test. Results: Growth, acid production, and adhesion of S. mutans were inhibited in all experimental groups. Expression of gft and fructosyltransferase in S. mutans was inhibited in all groups. A mixture of NaF and BS significantly reduced growth, acid production, adhesion, and gene expression of S. mutans compared with the other groups. Conclusion: Results of the present study demonstrated that a mixture of NaF and BS was useful as a mouth rinse in preventing dental caries.

Objectives: As suppression of mutans streptococci in young children may prevent or delay colonisation of the oral cavity, tooth brushing with dentifrices containing anti-Streptococcus mutans activity may aid in preventing caries. The aims of this study were to compare the effects of children's toothpastes, mouth rinses, oral antiseptics and fluoride solutions on the growth of Streptococcus mutans and non-mutans bacteria (Streptococcus sanguinis and Lactobacillus acidophilus). Methods: The agar diffusion assay at neutral pH was used to determine bacterial growth inhibition. Zones of bacterial inhibition were measured using a micrometer gauge. Results: Dentifrices containing 1,450 ppm fluoride produced greater growth inhibition of both S. mutans and S. sanguinis than those with < 500 ppm. No inhibition was seen for pure solutions of sodium fluoride or sodium monofluorophosphate at fluoride concentrations up to 10%. Stannous fluoride exerted anti-bacterial effects at concentrations above 1%. Significant growth inhibition of both S. mutans and S. sanguinis was seen with sodium lauryl sulphate at 0.25% and with triclosan at 0.01%. The mouth rinses containing 2% chlorhexidine gluconate, 0.05% cetylpyridinium chloride and 0.05% sodium fluoride produced anti-bacterial effects against S. mutans, S. sanguinis and L. acidophilus. Of the pure compounds, 0.01% chlorhexidine produced the greatest zone of growth inhibition against S. mutans. Of the combinations tested, 0.1% sodium fluoride with 5% povidone iodine produced synergistic anti-bacterial effects against S. mutans and S. sanguinis. The combination of 10% povidone iodine with 0.5% sodium hypochlorite produced additive anti-bacterial effects against L. acidophilus. Interference was seen in some combinations such as chlorhexidine with sodium lauryl sulphate, most likely through anion-cation reactions. However, 0.1% sodium fluoride when combined with 0.01% chlorhexidine did not interfere with the anti-bacterial effects of chlorhexidine alone 3 against S. mutans or S. sanguinis, but it reduced the anti-bacterial effects of cetyl pyridinium chloride alone against these bacteria. Conclusion: Sodium lauryl sulphate and triclosan are major bacterial inhibitory compounds in children's dentifrices. Mouth rinses containing chlorhexidine, sodium fluoride and cetyl pyridinium chloride have growth inhibitory effects against Streptococcus mutans, Streptococcus sanguinis and Lactobacillus acidophilus. The combinations of povidone iodine with sodium hypochlorite and povidone iodine with sodium fluoride produced additive and synergistic effects respectively.
