Combined effect of propranolol, vincristine and bevacizumab on HUVECs and BJ cells

Abstract: Infantile hemangioma is one of the most common benign tumors affecting children, with ~10–15% requiring medical treatment. These tumors consist of endothelial cells and stromal components, including fibroblasts, pericytes and mast cells. Effects of propranolol treatment in combination with bevacizumab or vincristine on cell growth were compared in the current study using human umbilical vein endothelial cells (HUVECs) and BJ human normal fibroblasts (BJs) to determine potential synergic effects in vitro. Inhib… Show more

“…To investigate the role of SPDEF and SOX4, and their downstream genes, we performed experiments in HUVECs. HUVECS have been commonly used in vitro in experiments to investigate IHs and other vascular tumors or angiogenesis in tumors [31,32]. The current studies report that the androgen-responsive gene SPDEF is mainly related to tumors -SPDEF was reported as a suppressor in tumor metastasis and progression, while some studies report otherwise [33].…”

“…Bevacizumab and propranolol have already been used as solutions in the same dosage as the ones employed in the current study [ 15 , 30 ]. Regarding a possible negative interaction between drugs, there is a previous report of synergistic effects of propranolol and bevacizumab, in vitro, to inhibit the growth of human umbilical vein endothelial cells (HUVECs) and BJ human normal fibroblasts (BJs) [ 31 ].…”

“…O BVZ e o propranolol já foram utilizados separadamente em forma de soluções na mesma dosagem empregada neste estudo 34,72 . Em relação a possível interação negativa entre drogas, há um relato prévio de efeitos sinérgicos de propranolol e BVZ, in vitro, para inibir o crescimento de células endoteliais da veia umbilical humana (HUVECs) e fibroblastos humanos normais (BJs) 73 .…”
