Combined Angiotensin II Type 1 and Type 2 Receptor Blockade on Vascular Remodeling and Matrix Metalloproteinases in Resistance Arteries

Abstract: Abstract-We investigated the role of angiotensin II type 1 (AT 1 ) and AT 2 receptors, matrix metalloproteinases (MMPs), and extracellular matrix (ECM) components involved in vascular remodeling of resistance arteries induced by angiotensin II (Ang II). Sprague-Dawley rats received Ang II (120 ng/kg per minute SC) Ϯ the AT 1 antagonist losartan (10 mg/kg per day PO), the AT 1 /AT 2 antagonist Sar 1 -Ile 8 -Ang II (Sar-Ile; 10 g/kg per minute SC), or hydralazine (25 mg/kg per day PO) for 7 days. Structure and m… Show more

“…Furthermore, the activation of 4E-BP1 in mesenteric arteries could be associated with vascular remodeling, which is not observed in aorta. 41 This suggests that PPAR-␥ can exert an inhibitory effect on vascular remodeling in type 2 diabetic and hypertensive patients, because type 2 diabetic patients exhibit significant remodeling of small arteries compared with nondiabetic normotensive patients. 42 It is well known that cell growth is under the control of many kinases and phosphatases.…”

“…Irbesartan, an angiotensin II receptor antagonist, inhibits MMP activity in patients with symptomatic carotid artery stenosis [114]. Inhibiting both angiotensin II type 1 and type 2 receptors with Sar1-Ile8-Ang II reduces collagen type I and elastin deposition but also increases vascular stiffness, fibronectin, and MMP-2 activity [115]. Olmesartan improves LV remodeling in apo E null mice by inhibiting nuclear factor -B (NF-B) and MMP-9 activities [116].…”

“…The ECM content, such as collagen and elastin in the walls of arteries, is increased in hypertensive patients and in genetic or experimentally-induced hypertension in animals (5,6). Many researchers have studied the mechanisms of collagen accumulation during hypertension.…”
