Combination Treatment with Resveratrol and Sulforaphane Induces Apoptosis in Human U251 Glioma Cells

Abstract: Resveratrol is a naturally occurring polyphenolic compound highly enriched in grapes, peanuts, red wine, and a variety of food sources. Sulforaphane belongs to the family of isothiocyanates and is highly enriched in cruciferous vegetables. Our previous study showed that resveratrol, when used at high concentrations, inhibited cell proliferation, caused the cell cycle arrest and induced apoptotic cell death in glioma cells. In the current study, we tested the effect of combination treatment with resveratrol and… Show more

“…For example, the administration of sulforaphane and resveratrol (polyphenolic compound) inhibited cell proliferation and migration, reduced cell viability, induced lactate dehydrogenase release, decreased pro-survival Akt phosphorylation, and increased caspase-3 activation in human U251 glioma cells (126). A combination of sulforaphane and the flavonoid apigenin was shown to modulate gene expression of phase II detoxifying enzymes (including GST and UGT) in the human epithelial colorectal adenocarcinoma cell line, Caco-2 (275).…”

“…Another consideration that we have to take into account is that other antioxidants compounds of red wine could give more stability to the resveratrol than the pure resveratrol 56,57 . This could be the reason why resveratrol at µM concentration is currently used in the literature [58][59][60][61] . There are few reports of the effects of red vs white wine on plasma SHBG levels: a 5% reduction in plasma SHBG levels was observed in premenopausal drinking red wine vs white wine, but the red wine used in that study contained only 0.9 mg/L of resveratrol 62 .…”

“…RES is a naturally-occurring compound that exhibits cytoprotective, antioxidant and anti-inflammatory vasoprotective properties 31 . Most studies examining the mechanism of action of RES proposed that preventing and/or inhibiting the development of cardiovascular diseases is related to its ability to reduce oxidative stress and associated inflammation and thus, ameliorate diseases that become more common with aging, such as hypertension 23,31 .…”

For example, the consumption of 25 mg kgG 1 RES in drinking water for 14 days reduced for 7 weeks, control rats did not receive RES oxidative stress, improved endothelial functions and protected rats against development of pulmonary hypertension 23 .…”
