Colorimetric sensing strategy for heparin assay based on PDDA-induced aggregation of gold nanoparticles

Abstract: We have developed a highly sensitive colorimetric sensor for the detection of heparin based on its anti-aggregation effect of the PDDA–gold nanoparticle colloidal system.

“…The analyte-triggered aggregation for chemical and biological sensing applications is performed based on the affinity of analytes such as the electrostatic or hydrogen-bonding interaction toward unmodified AuNPs. For example, Ma et al devised a simple assay for the sensitive and selective colorimetric detection of heparin [76]. In the absence of heparin, the poly(diallyldimethylammonium chloride) (PDDA) with positive charges can easily adsorb onto the citrate-capped AuNP surfaces by electrostatic attraction (Figure 7).…”

“…However, when heparin is present, the PDDA molecules will interact with the heparin, which prevents the PDDA-induced aggregation of the AuNPs. Reproduced with permission from Reference [76]. Royal Society of Chemistry, 2019.…”

“…[6][7][8]31,32] A ratio frequently used in this type of study is calculated as LSPR maximum absorptions of the non-aggregated versus aggregated nanoparticles, Abs Non-Agg /Abs Agg . [8,31,[33][34][35][36][37][38] For instance, these types of ratios were previously used for ≈14 and ≈35 nm spherical AuNPs as Abs 525nm /Abs 585nm and Abs 542nm / Abs 700nm , respectively. [31,33] Based on UV-vis spectra from the examples in Figure S8, Supporting Information, a higher ratio is expected for the Au nanoprobe hybridized with a fully complementary DNA compared to hybridization with a mismatched DNA, corresponding to differences in their aggregation profile.…”

“…Colorimetric sensors produce colorimetric signal induced by a coloration reaction of catalyst, chromogenic substrates and H 2 O 2 , [49] which is reliable, easy operational and less time-consuming. [120] Biocatalytic MOF-based colorimetric sensors shows emerging application in biomarker detection. For example, to screen α-Glucosidase (GAA)…”
