Colletotrichum gloeosporioides lipase: Characterization and use in hydrolysis and esterifications

Abstract: Colletotrichum gloeosporioides was evaluated for its capacity to produce extracellular lipase. A crude enzyme preparation obtained after 48 h of fermentation reached 742 U/L of lipolytic activity. Estimated molecular weight of proteins responsible for this activity was about 18 kDa as determined by SDS-PAGE and zymogram analysis. Enzyme preparation showed optimum pH at 10 and stability at optimum temperature (37°C) for 5 h. It showed tolerance to a wide range of salts (NH 4 + , Mg +2 , Ca +2 , Mn +2 and Sn +2)… Show more

“…Nutritionally the palmitic and stearic acids are the most commonly consumed saturated fatty acids in the Western diet, each having different effects on fasting serum lipoproteins (van Rooijen et al, 2021). Unsaturated fatty acid levels are generally higher than saturated ones in mushrooms (Sande et al, 2015). A stereoisomer of vaccenic acid (cis-VA), which is an omega-7 fatty acid (ω-7), showed an 11% abundance in our results.…”

“…Additionally, two unsaturated omega fatty acids, the methyl esters 9,12-octadecadienoic acid (omega-6, ω-6) and 9-octadecenoic acid (omega-9, ω-9) were reported as being present. Omega-6 is an essential fatty acid present in most mushroom species, playing a vital role in many physiological functions, such as maintaining bone health, regulating metabolism, and stimulating skin and hair growth (Sande et al, 2015;Al-Khudairy et al, 2015). Omega-9 is a monounsaturated fatty acid typical in vegetable oils, known for its effectiveness in reducing cholesterol levels (Puiggros et al, 2002).…”

“…Esterification reactions are often conducted in the presence of non-absorbable organic solvents and under circumstances of low water content. This choice of reaction medium is favored due to its ability to enhance substrate solubility [40].…”

“…The developed method was also successfully applied to unconcentrated crude microbial culture media. We were able to detect lipolytic enzymes produced by Aspergillus aculeatus [ 29 ], Colletotrichum gloeosporioides [ 30 , 31 , 32 ], Fusarium solani [ 33 , 34 , 35 , 36 , 37 , 38 ], Penicillium expansum [ 6 , 39 , 40 , 41 , 42 ], and Trichoderma harzianum [ 43 , 44 , 45 , 46 , 47 ]. The choice of substrates used in chromogenic agar detection is not limited to triglycerides, but could also extended to di- or monoglycerides, vinyl laurate and other acyl ester substrates, which would be beneficial for screening of partial-glyceride lipases or steryl esterases.…”
