Clotting Predisposition in Carcinoma of the Prostate

Abstract: To delineate factors that might increase the risk of thromboembolic events in patients on estrogen therapy for prostatic carcinoma a study was undertaken of various coagulation parameters. A correlation was demonstrated between advanced stages of prostatic carcinoma and depression of the serum antithrombin III activity. The addition of estrogen therapy resulted in further depression. Since low levels of serum antithrombin III activity have been associated with thrombosis this observation may have clinical rele… Show more

“…Coagulation abnormalities in cancer patients are well documented, and a hypercoagulable state leading to vascular thrombosis has been reported (Miller et al, 1967;Davis et al, 1969). Attempts have been made to study coagulation abnormalities following various therapeutic modalities in various cancers, particularly with reference to chemotherapy (Hicsonmez and Buyukpamukcu, 1977;Dobbs et al, 1980;Enck and Rias 1984;Goodnough et al, 1984). The influence of cytotoxic agents on coagulation factors and platelet function has not so far been well defined, and published data are conflicting (Canobbio et al, 1986).…”

“…In a recent study (Dobbs, Barber, Weigel & Bergin, 1980) a correlation between advanced stages of prostatic cancer and depression of serum antithrombin 111 activity was demonstrated. The addition of estrogens resulted in a further depression of antithrombin 111.…”

“…DES also decreases fibrinolysis by reducing levels of plasminogen activator [49]. It has also been noted that another important factor in the coagulation cascade, antithrombin III, is significantly decreased with oestrogen administration [50,51]. Other proposed mechanisms through which oestrogens contribute to cardiovascular complications include alterations in platelet function and serum lipid profiles [52,53].…”
