Cloning, partial sequencing and expression of a cDNA coding for branching enzyme in cassava

Abstract: Branching enzyme is involved in the synthesis of amylopectin in plant reserve starch. A cDNA coding for cassava (Manihot esculenta Crantz) branching enzyme was cloned from a lambda gt11 cDNA library using a potato cDNA probe. The cloned cDNA was partially sequenced. The sequence data confirmed the identity of the clone when compared to that of potato, the homology being ca. 80% at the nucleotide level and 85% at the amino acid level. Furthermore, the cloned cassava cDNA was able to restore branching enzyme act… Show more

“…The observed tissue specific expression pattern of Meisa1 was similar with previously reported patterns from other starch metabolizing genes in cassava (Salehuzzaman et al, 1992;Salehuzzaman et al, 1994;Baguma et al, 2003). Meisa1 declined with increased developmental stage of the storage root (Fig.…”

“…The occurrence of two or more copies of Meisa1 and GbssI genes could be due to the ploidy level of cassava genome (Awoleye et al, 1994). However, SbeI (Salehuzzaman et al, 1992) and SbeII (Baguma et al, 2003) have been reported to occur as single copies. These two genes could also be a consequence of duplication of the same ancestral gene.…”

“…Genomic and eDNA libraries from leaves, roots, and cotyledons of cassava have been constructed (Hughes et al, 1992;Salehuzzaman et al, 1992;Tenjo and Mayer, 1993;Bohl et al, 1997). These can be used to isolate genes and promoters from cassava, and also to characterize cassava germplasm (Beeching et al, 1993).…”
