Cloning and sequencing the endocarditis immunodominant antigen of Streptococcus sobrinus strain MUCOB 263

Abstract: Summary.Immunoblotting sera from cases of Streptococcus mutans or S. sobrinus endocarditis against an extract from S. sobrinus strain MUCOB 263 had identified three immunodominant antigenic bands at 190,200 and 220 kDa. A lambda ZAPII DNA library was produced from the sheared genomic DNA of S. sobrinus MUCOB 263 and six identical positive clones were identified when this library was screened with serum from a patient with endocarditis caused by a bacterium from the mutans group of streptococci. On subcloning a… Show more

“…Antibodies to surface PA were elevated greatly in patients with S. mutans infective endocarditis (24). Antibodies against similar surface proteins were reported in S. sobrinus (25), and S. oralis (26). In S. mutans , including the untypable strains used here, PA gives a common immunological specificity and is highly immunogenic.…”

“…Cellular proteins of 0.5 ml of denatured cell suspension were separated by SDS–PAGE using an 8 cm wide sample slot and Western‐blotted onto nitrocellulose (Mini‐Protean II gel apparatus, Bio‐Rad). After blocking, incubation with 20 ml of 1:400 diluted serum PRT and washing, bound antibody was eluted from the membrane [17]. Duplicate Western blots of E. coli XLOLR/pSsaA and cellular and supernatant proteins recovered from both S. epidermidis NCTC 11047 and S. aureus NCTC 6571 were immunoblotted with serum PRT or the eluted anti‐SsaA antibody.…”

“…Dental caries is one of the most prevalent infectious diseases in humans [1,2] and has been associated with major clinical complications, such as infective endocarditis [3][4][5]. The World Oral Health Report 2003 [6], published by the World Health Organization, indicates that dental caries is a major health problem in most industrialized countries, affecting children, adolescents, and adults.…”
