Cloning and sequence analysis of complementary DNA encoding a precursor for chicken natriuretic peptide

Abstract: Chicken ¢c.natriuretlc p~ptide (=.¢hNP) has hcen identified in ehteker~ heart, which allowed hi=her ha=nolo=:/to brain natriuretic peptide (BNP) than to atrial natrturetic peplide (ANP) [1]. Complementary DNA (eDNA} clone en~odir~j a chNP pr~ursor (pre-chNP) was isolated from ~rdiac eDNA library mtd sequenced. Pre.ehNP was 140.residue pepttde carrying a 24.residue fiMnal peptide at the N-tennlnus and a.ehNP at the C.termi. nus, and did not exhibit high homolosy to porcine BNP except for the C.terminal re=ion. … Show more

“…Since BNP is the only natriuretic peptide that has not been identified in eels, VNP could be defined as novel if a distinction between VNP and BNP could be clarified. It has been shown that all BNP cDNAs from mammals and birds so far reported invariably have seven repetitive ATTTA sequences in the 3' non-coding region (Kojima et al 1989;Akizuki et al 1991), which is known to accelerate mRNA degradation (Wilson & Treisman, 1988). Thus, the cDNA sequence of VNP could provide more definite information on the distinction between VNP and BNP.…”

“…However, the cDNA of eel VNP lacks the seven repetition of ATTTA sequence in the 3' non-coding region (Takei Y. and Nishizawa T., unpublished data). The presence of the repetitive sequence is a characteristic of all BNP cDNAs so far sequenced (Akizuki et al 1991). It remains to be determined whether the fourth natriuretic peptide, namely BNP, is present in the eel or not.…”

“…We note that the timing of Fig. 1 Alignment of the atrial natriuretic factor predicted protein sequence from Xenopus, Rana (D01043), human (Nemer et al 1984) and chick (Akizuki et al 1991). The first 23 amino acids of Xenopus ANF (overlined) correspond to the signal sequence.…”
